抗人绒毛膜促性腺激素单克隆抗体的研制与鉴定

摘　　要：利用常规杂交瘤技术制备了２株抗人绒毛膜促性腺激素（ｈＣＧ）的杂交瘤细胞。ＥＬＩＳＡ结果表明，单克隆抗体ＩＤ５和４Ｄ５均与人ｈＣＧ有特异性反应，腹水效价分别为３．２×１０－５、６．４×１０－５，均属于ＩｇＧ１亚类。与人促卵泡激素（ｈＦＳＨ）和人促黄体激素（ｈＬＨ）的交叉反应呈阴性。免疫转印结果显示能识别相对分子质量为２４×１０３ｕ的ｈＣＧ－β链条带。亲和常数分别为１．０１×１０７Ｌ／ｍｏｌ、４．４３×１０７Ｌ／ｍｏｌ。Two hybridomas named as 1D 5 and 4D 5 stably producing monoclonal antibody (McAb)against human chorionic gonadotrophin (hCG)were established using the routine hybridoma technique.It is found that these McAbs reacted specially with hCG β but not with human luteinizing hormone(hLH) and follicle stimulating hormone (hFSH).Both of them belonged to mouse IgG1,and the ascitic titres were 3.2×10 -5 (1D 5)and 6.4×10 -5 (4D 5)respectively.The immunoblotting results showed that both 1D 5 and 4D 5 recognized the band of hCG β chain with a molecular weight of 24×10 3u.The affinity constant for the McAbs were 1.01×10 7 L/mol and 4.43×10 7 L/mol respectively by ELISA.
