摘  要:目的检测脑型脂质运载蛋白前列腺素D合成酶(LPGDS)和前列腺素D受体(DP)在早孕期小鼠子宫中的表达变化,评价LPGDS和DP在胚胎着床和妊娠维持中的作用。方法用半定量逆转录聚合酶链反应(RT-PCR)的方法检测LPGDS和DP在孕1~8d小鼠子宫中的表达水平,以及在胚胎着床和非着床位点(妊娠第5天)的表达。结果小鼠孕2~4d,LPGDS mRNA的水平明显高于孕1、5~8d;孕5~8d,DP mRNA的水平显著高于孕1~4d。孕第5天,胚胎着床和非着床位点LPGDS的表达没有差异,DP在胚胎非着床位点的表达显著高于着床位点。结论LPGDS在小鼠胚胎着床前子宫中高表达,DP在胚胎着床后高表达;说明LPGDS可能与子宫发育成为可接受态有关,而DP作用在于维持妊娠。Objective: To investigate the expression profile of lipocalin-prostaglandin D synthase (LPGDS) and prostaglandin D receptor (DP) in mouse uterus during early pregnancy to observe their roles in the implantation and maintenance of pregnancy. Methods: Semi-quantitative RT-PCR was used to detect the expression of LPGDS and DP in mouse uterus from day 1 (D 1) to day 8 (D 8) of pregnancy, and their expressions in the implantation site and interimplantation site on D5 were compared. Results:The LPGDS mRNA levels from D 2 to D 4 were significantly higher than those on D 1, D 5, D 6, D 7 and D 8 (P〈0.01) ; the DP mRNA levels from D 5-D 8 were significantly higher than those from D 1-D 4 (P〈0.01). No significant difference was found in the LPGDS mRNA levels between the implantation site and inter-implantation site on D 5 of pregnancy, but DP mRNA levels in the inter-implantation site were higher than those in the implantation site. Conclusions:The high expression of LPGDS in pre-implantation uterus suggests that LPGDS may be involved in the preparation of uterus for embryo implantation. Higher expression of DP after embryo implantation may play an important role in the maintenance of pregnancy.
