离体毛囊切片两种制作方法的比较及凋亡检测

摘　　要：目的对人头皮游离毛囊(HF)切片制作及凋亡检测方法进行比较。方法分别将石蜡切片法和冰冻切片法应用于游离毛囊;利用原位缺口末端标记法(TUNEL)进行荧光染色与3,3′-二氨基联苯胺(DAB)染色,检测未经培养的游离毛囊及培养5 d的游离毛囊的不同部位凋亡细胞情况。结果在对游离毛囊形态检测中,各组毛囊在外根鞘(ORS),毛球(HB)下部,真皮鞘(DS)及毛乳头(DP)中的凋亡细胞数并无显著性差异(P>0.05)。结论冰冻切片法应用于游离毛囊简单有效,TUNEL原位荧光结合DAB染色适合于检测毛囊细胞凋亡情况。Objective To compare two methods of section making and to detect the apoptosis on human hair follicle（HF） in vitro. Methods The paraffin section and frozen section methods were used. The TUNEL [fluorescence staining and 3, 3＇-Diaminobenzidine（DAB） staining] was used to investigate the apoptotic cells of different parts when the hair follicles were uncultured and cultured on 5th day. Results The process of making frozen section was more simple and feasible on single hair follicle. The apoptotic index of outer root sheath（ORS）, lower part of hair bulb（HB）, the dermal sheath（DS） and dermal papilla（DP） of every groups had no difference （P〉0.05）. Conclusion The method of frozen section was more feasible on the section making of single hair follicle. The TUNEL with fluorescence staining and DAB staining was suitable for the detection of apoptotic status of hair follicle.
