摘  要:以制备适用于疫苗生产检定中病毒鉴别试验和外源因子检查的高效价腮腺炎病毒抗血清为目的。用腮腺炎病毒接种SPF鸡胚尿囊腔,培养收取病毒尿液免疫SPF鸡,采集抗血清。腮腺炎病毒接种Vero细胞,培养病毒抗原经PEG沉淀,超速离心法纯化后免疫家兔采集抗血清。比较两种免疫方法所得病毒抗血清效价。结果显示SPF鸡抗腮腺炎病毒血清中和抗体GMT为1:1716,兔抗腮腺炎病毒血清中和抗体GMT为1:732。两种动物抗血清均适用于疫苗生产相关检定。免疫SPF鸡制备的病毒抗血清无特定病原及抗体污染,是毒种外源因子检测和疫苗鉴别试验的理想试剂。免疫SPF鸡制备病毒抗血清的程序简单,结果易于验证,有利于生物试剂标准化。Preparation of high titer at, antisera for mumps virus identification and virus neutralization test for quality control in the vaccine production. SPF chickens were immunized by mumps virus propagated in embryonating eggs from a SPF flock and rabbits immunized by the virus propagated in Vcro cells , concentrated by PEG precipitation and purified by ultra centrifugation. Both sera were compared, the cumulative titcr of antisera from SPF chikens for neutralization antibodies was 1 : 1716 in GMT, the antisera from, rabbits had a result of 1:732 in GMT. Both sera are suitable for qualities control of the mumps vaccine either monovalence or multiplevalence as in MMR or measles-rubella combined vaccine. The antisera prepared from SPF chickens have been free from antibodies contamination to related pathogens, it is essential for quality control of vaccine. The antisera are easy to prepare and the result could be directly proved.
