摘 要:对15例良性前列腺增生(BPH)和12例正常前列腺组织标本中胰岛素样生长因子-I-(IGF-I)mRNA的表达情况进行了研究。RT-PCR的方法可自所有标本中检出IGF-ImRNA的表达,并用电泳和Southern杂交证实了结果的可靠性。原位杂交表明13/15的BPH和9/12的正常前列腺标本中有IGF-ImRNA表达,IGF-I在上皮的表达强于间质,尤以基底细胞表达最明显。IGF-ImRNA在BPH组织中的量多于正常前列腺。In situ hybridization and reverse transcription PCR were adopted to map the insulin-like growth facto-I(IGF-I)gene expression in 15 benign hyperplastic and 12 normal prostatic tissues. IGF-I mRNA could be detected by RT-PCR in all the tissue samples,With in situ hybridization,13 of the 15 benign hyperplastic prostatic tissues and 9 of 12 normal prostatic tissue expressed the IGF-I mRNA. The latter was mainly located in the epithelial cells especialy in the basal cells and was expressed at low levels in the stromal cells,The level of expression in BPH was much higher than that in normal prostate suggesting that IGF-I might play an important role in the growth regulation of prostate and the pathogenesis of BPH.
