摘 要:目的:观察骨髓干细胞移植对正常和心肌梗死大鼠心电图的影响及其治疗作用。方法:实验于2003-03/2004-03在哈尔滨医科大学附属第一医院动物研究室完成。取自哈尔滨医科大学附属第一医院饲养的Wister大鼠作为实验对象,体质量(50±10)g幼鼠20只做为干细胞的来源,将体质量(300±50)gWister雄性大鼠80只作为细胞移植的受体,并将其随机分成4组,每组20只。①正常对照组:用1mL注射器取200μL细胞培养液基质移入到正常心肌组织中。②心肌梗死对照组:取200μL细胞培养液基质移入到心肌梗死大鼠瘢痕组织中。③正常移植组:在正常心肌组织中移入200μL干细胞悬液。④心梗移植组:在心肌梗死后瘢痕组织中移入干细胞悬液。通过结扎左冠状动脉3周形成心肌瘢痕模型。观察骨髓干细胞移植前、移植后即刻和饲养4周的心电图变化。结果:80只实验大鼠中,在心肌梗死模型制作和细胞移植过程中死亡39只,最终进入结果分析大鼠41只,其中心肌梗死对照组7只,心肌梗死移植组8只,正常对照组和正常移植组各13只。①幼鼠骨髓干细胞经体外培养、5-溴脱氧尿核苷标记和5-氮杂胞嘧啶核苷诱导能形成幼稚的心肌样细胞。将其移植到大鼠正常心肌组织和心肌梗死后的瘢痕心肌组织中4周后发现不同心脏组织环境中能生存增殖良好,替代坏死心肌。②正常对照组大鼠心电图显示Ⅱ,Ⅲ,avF主波向上,avR主波向下,心率为(300±50)次/min。心肌梗死对照组QRS波增宽,ST段抬高,可出现各种心律失常。正常移植组和心肌梗死移植组大鼠均表现为心律不稳,ST段下移或抬高等一过性缺血或损伤性改变;细胞移植后4周,正常移植组和心梗移植组大鼠均未见有害的心律失常出现,而且,心肌梗死移植组大鼠与心肌梗死对照组(或移植前)相比QRS波变窄,R波相对升高,ST段回到基线。结论:骨髓干细胞移植到心肌梗死后�AIM: To observe the effect of tnmsplantation of bone marrow stern cells on the electrocardiogram (ECG) of normal and myocardial infarction rats and its therapeutic effect. METHODS: The experiment was conducted at the Animal Institute of First Hospital Affiliated to Harbin Medical University from March 2003 to March 2004. Wistar rats, which were raised in the First Hospital Affiliated to Harbin Medical University, were used for experimental subjects. 20 Wister young rats with the body mass of (50±10)g were selected as donor for cellular transplantation and 80 male Wister rats with the body mass of (300 ±50)g were selected as the receptor of cell transplantation and randomly divided into 4 groups with 20 rats in each group: ① Normal control group: 200μL cell culture medium was injected into the normal myocardial tissue by using a 1 mL tuberculin syringe . ② Myocardial infarction control group: 200 μL Cell culture medium was injected into the scar tissue of the myocardial infarction rats. ③ Normal transplantation group: 200μL stem cell suspension was injected into the normal myocardial tissue. ④ Myocardial infarction transplantation group : Stem cell suspension was injected into the scar tissue after myocardial infarction. Myocardial scar model formed 3 weeks after ligation of left coronary artery. ECG change was observed before, immediately after and 3 weeks after transplantation of bone marrow stem cell. RESULTS: Among 80 experimental rats, 39 died in the preparation of myocardial infarction model and the process of cell transplantation, finally, 41 rats entered the stage of result analysis. Among them, 7 rats were in the myocardial infarction control group, 8 in the myocardial infarction transplantation group and 13 in the normal control group and normal transplantation group separately. ① Bone marrow stem cells of young rats can form immature myocardial-like cells after in vitro culture ,labeling of 5-bromodeoxyuridine and induction of 5-azacytidine.
