摘 要:目的 了解健康中国女性个体X染色体失活偏移(SXCI)的发生状况,明确年龄因素是否与之关联。方法 采集401例健康中国女性的外周血标本,提取基因组DNA,经HpaⅡ消化后,PCR扩增雄激素受体(AR)基因,产物经变性聚丙烯酰胺凝胶电泳,银染显示结果,比较HpaⅡ消化前后等位基因扩增产物的强度差异,计算其校正比值(CR)。结果 无论采用CR≥3抑或≥10作为标准,老年组(50岁~96岁)SXCI发生率均显著高于新生儿组(出生后4周以内)或青年组(16岁~50岁),而新生儿组与青年组之间差异不显著。采用标准CR≥3时,新生儿组为4.3%(P=0.532vs青年组),青年组为7.8%(P=0vs老年组),老年组为25.7%(P=0.001vs新生儿组)。采用CR≥10标准时,新生儿组为2.2%(P=1vs青年组),青年组为1.2%(P=0vs老年组),老年组为11.1%(P=0vs新生儿组)。结论 健康中国女性SXCI发生率是随年龄的增长而变动的,年龄与女性个体SXCI的发生相关。在筛选女性肿瘤易感个体时,应注意用作对照的健康女性的SXCI的发生状况。Objective To investigate whether age is associated with X- chromosomal inactivation skewing in blood cells in Chinese healthy females . Methods DNA was extracted from mononuclear blood cells from 401 Chinese healthy females. Exon 1 of androgen receptor (AR) gene was amplified, with its products from different alleles resolved on denaturing polyacrylamide gels and visualized by silver staining. Corrected ratio ( CR ) of the AR allele for each HpaⅡ digested and auto - control sample pair was calculated. All statistical tests were two - sided. Results Comparisons were made among three age groups: ① neonates, ②young women group of females 16 to 50 years old and, ③old women group of females 50 to 96 years old. With CR≥3 adopted as the criterion, the incidence of skewing was 4.3% ( 2 of 46) in neonates(P =0. 532 vs young women group) ,7.8% ( 13 of 166) in young women group(P = 0vs old women group), and 25.7% ( 37 of 144) in old women group (P = 0. 001 vs neonates). When a more stringent criterion of CR≥ 10 for skewing was used, the incidence of skewing was 2.2% ( 1 of 46) in neonates (P=1vs young women group) ,1.2% ( 2 of 166) in young women group(P =0vs old women group) ,and 11.1% ( 16 of 144) in old women group(P=0vs neonates). Conclusion Age is associated with X -chromosomal inactivation skewing in blood cells in Chinese healthy females. More attention should be paid to X - chromosomal inactivation skewing in blood cells in Chinese healthy females screened for the ones predisposed to cancer.
