摘  要:目的探讨恶性疟原虫AMA-1(Ⅲ)重组蛋白的免疫活性。方法利用毕赤酵母高效表达系统分泌表达AMA-1(Ⅲ),表达产物用纯化后免疫新西兰兔,检测其血清中IgG滴度的变化,观察重组AMA-1(Ⅲ)免疫效应。结果重组AMA-1(Ⅲ)免疫后,ELISA检测新西兰兔血清中IgG滴度发现在3次免疫后OD490值明显高于佐剂免疫对照组(P<0.01)。结论AMA-1(Ⅲ)重组蛋白具有较好的免疫原性。Objective To investigate the immunogcnicity of recombinant Plasrnodium falciparum apical membrane antigen 1 C-terminal domain [Pf AMA-1 (Ⅲ) ] expressed in Pichia pastoris. Methods Recombinant Pf AMA-1 (Ⅲ) were successfully expressed in Pihia pastoris and purified with Ni-NTA column. Rabbit was immunized with recombinant Pf AMA-1 (Ⅲ) formulated with Freund's adjuvant subcutaneously 3 times at three week intervals. After immunization, kinetic of antibody response was determined by ELISA. Result Significant immuneresponses were induced in New Zealand rabbits (detected with ELISA) after the third immunization. Conclusion Significant immune response was induced in New Zealand rabbits by the recombinant Pf AMA-1 (Ⅲ).
