环胞霉素A对一氧化氮生成作用的影响

摘  要:目的研究环胞霉素A对一氧化氮生成作用的影响。方法培养血管内皮细胞ECV304,分为TNFα组和TNFα+CsA组,TNFα刺激浓度为50ng/mL,CsA浓度为0.1、1、10μmol/L,培养24h后测定上清中的NO浓度;临床研究中的20例膜性肾病患者分为治疗组14例和对照组6例,治疗组服用CsA5mg/kg/d和强的松1mg/kg/d,对照组强的松1mg/kg/d,治疗4周,分别测定治疗前后外周血中的NO含量和24小时尿蛋白量。结果培养细胞研究发现TNFα组NO为74.63±5.83μmol/L,TNFα+CsA(0.11、、10μmol/L)组依次为11.49±1.72μmol/L9、.78±1.05μmol/L9、.54±0.83μmol/L,二者之间存在明显差异,P<0.01;14例膜性肾病患者治疗前后血NO浓度分别为197.58±21.64μmol/L、125.39±16.46μmol/L,二者之间存在明显差异,P<0.01,而对照组治疗前后为186.72±15.43μmol/L和179.45±18.32μmol/L,没有明显差别,治疗组4周前后的24小时尿蛋白量分别为4.53±0.92g和3.81±1.26g,P<0.01,而对照组前后变化无统计学意义。结论环胞霉素A可以抑制TNFα诱导的NO产生。Objective To investigate the effects of cyclosporin A on nitric oxide synthesis induced by TNF1^1A. Method Cultivated ECV304 cells were stimulated by TNF α (50ng/mL) or TNF α (50ng/mL) plus CsA(0.1,1,10μ mol/L).NO metabolites of supernatant was detected.ln clinical study fourteen patients with membranous nephropathy accepted four weeks treatment of cyclosporin A(5mg/kg/day) and prednisone(1 mg/kg/day). Six controls with the same disease treated only with prednisone. Plasmal NO metabolites and the amount of total 24h urine protein around treatment were examined. Results The present results shows: the supernatant accumulation of NO metabolites was significantly decreased by CsA(0.1,1,10 μ mol/L),expectly from 74.63 ±5.83 μ mol/L to 11.49 ± 1.72 μ mol/L, 9.78 ± 1.05μmol/L, 9.54 ± 0.83μmol/L.P〈0.01. In vivo,placmal NO metabolites of patients treated with CsA was decreased from, 197.58 ± 21.64μ mol/L to 125.39 ± 16.46 μ mol/L,P〈0.01;the amount of 24h urine protein from 4.53 ± 0.92g to 3.81 ± 1.26g,P〈0.01. The controls had no significant changes. Conclusion NO synthesis in response to TNF1^1A can be inhibited by CsA.
