摘  要:目的探讨人参总皂甙对大鼠缺血再灌注脑组织的保护作用机制。方法利用大鼠大脑中动脉闭塞模型,脑组织切片行Nissel和TUNEL染色,计算调亡的细胞数。结果缺血非治疗组神经细胞数量明显减少,部分神经细胞胞体皱缩,核固缩,具有凋亡细胞的某些光镜特征。治疗组仅见少量神经细胞变性,胞体变形缩小。结论人参总皂甙能明显抑制神经细胞凋亡,从而发挥其神经营养和神经保护作用。Objective To study the protective effect of ginseng total saponin on the brain tissue in isehemia-reperfusion rat model. Methods 40 adult male Wistar rats were randomly divided into 5 groups: the normal group, The sham group, the isehemic-reperfusion group, ginseng total saponin- treatment group 1, ginseng total saponin- treatment group 2. The model of focal cerebral ischemia was made by occluding middle cerebral artery (MCA) for 2h and reperfusing for 48h. The rats received either ginseng total saponin (50 mg/kg of body weight. ) or saline ip just before the beginning of reperfusion and the brains were removed after 48h, and the ginseng total saponin -treated 2 rats reiceived ginseng total saponin for 3 days and their brains were removed after 72h. The brain tissues were used for the ob- servation of cells with the method of Nissle staining, TUNEL, and the data were calculated by using im age analysis system. Results In the ischemie group, The nummber of neurocyte reduced obviously. Parts of neurocyte atrophia. Nuclear atrophia , have some characters of apoptosis. In the ginseng total saponin -treated group, similar changes could also been seen but less than those observed in the ischemie group. Conclusions Ginseng total saponin play an important role in protecting neurons from ischemia-induced apoptosis.
