摘 要:目的观察中药复方制剂延肾(以下简称延肾制剂,YSH)对重组人白细胞介素-1β(rhIL-1β)刺激后体外培养的人肾小球系膜细胞产生细胞间粘附分子-1(Intercellular adhesion moleculeI,ICAM-1)和血管细胞粘附分子-1(Vascular celladhesion molecule,VCAM-1)的影响,冀以从细胞生物学水平探讨延肾防治慢性肾功能衰竭(Chronic renal failure,CRF)的作用机制。方法采用双抗夹心ELISA法,分别加入重组rhIL-1β及rhIL-1β+YSH大鼠血清。观察体外培养的人肾小球系膜细胞在rhIL-1β刺激后,ICAM-1和VCAM-1的生成及YSH对ICAM-1和VCAM-1生成的影响。结果rhIL-1β可明显刺激人肾小球系膜细胞产生ICAM-1和VCAM-1,YSH大鼠血清对rhIL-1β刺激后人肾小球系膜细胞所产生的ICAM-1和VCAM-1有抑制作用,其抑制程度与药物浓度有一定的量效关系。结论延肾对rhIL-1β刺激后肾小球系膜细胞所产生的ICAM-1和VCAM-1有明显的抑制作用,提示YSH抑制系膜细胞自分泌ICAM-1和VCAM-1是预防肾小球硬化发生发展的重要作用机制之一。Objective To study the effect of the traditional Chinese herbal medicine Yanshen(YSH) on the production intercellular adhesion molecules (ICAM- 1 ) and vascular cell adhesion molecules (VCAM- 1 ) by human mesangial cells (huMsC) cultured in vitro , YSH has the actions of expelling turbid - evil, invigorating qi and activating blood flow, The aim of this study was to explore the mechanism of YSH on prevention and treatment of chronic renal failure (CRF) in level of cell biology. Methods The effects of rhlL- 1β and rhlL- 1 β + YSH on the production of ICAM - 1 and VCAM - 1 by huMsC cultured in vitro were observed by using ELISA. Results ICAM - 1 and VCAM - 1 were significantly stimulated by rhlL- 1β in cultured huMsC in vitro , but were significantly inhibited by YSH in a dose- dependent manner. Conclusions YSH inhibits ICAM - 1 and VCAM- 1 produced by huMsC, which underlies the mechanism of preventing the occurrence and development of glomerulosclerosis.
