摘 要:脂质第二信使尤其磷酸肌醇在细胞信号途径网络中具有重要作用,3-磷酸肌醇激酶(PI3K)产生的特异肌醇脂质3,4,5-三磷酸肌醇和3,4-二磷酸肌醇是调节Akt活性的重要作用分子,与多种细胞功能有关。研究发现PI3K通过效应分子Akt能促进细胞增殖、抑制细胞凋亡,而且与癌症的发展有关。细胞核内自主性肌醇脂质代谢是细胞核内信号途径的重要成分。Lipid second messengers, particularly phosphoinositides, play a pivotal role in several cell signaling networks. Specific inositol lipids such as PtdIns(3,4) P2 and PtdIns (3,4,5)P3 which are generated by phosphoinositide 3-kinases (PI3Ks) are specific activators to the serine/threonine protein kinase Akt that have been implicated in a plethora of cell functions. Studies show that Akt stimulates cell proliferation and suppresses apoptosis, and is implicated in cancer progression. Many evidences have highlighted the presence of an autonomous nuclear inositol lipid metabolism, and suggest that lipid molecules are important components of signaling pathways in the nucleus.
