摘 要:目的研究血管活性肠肽(VIP)对感染时的leydig细胞分泌的细胞因子IL-1I、L-6和TGF-β的影响。方法SD大鼠的睾丸经Ⅱ型胶原酶消化、过滤及Percoll分离获得高纯度、高活率的leydig细胞,经不同剂量的VIP作用后,再感染溶脲脲原体(UU),观察在UU感染时,不同剂量VIP对leydig细胞分泌的IL-1(MTT法)I、L-6、TGF-β(ELISA法)的影响。结果UU感染时,VIP对leydig细胞分泌的细胞因子有明显的影响:能促进leydig细胞分泌IL-1(活性)、抑制IL-6(含量)分泌;同时既能促进(高剂量VIP)又能抑制(低剂量VIP)TGF-β分泌。结论在UU感染时,VIP能改变大鼠leydig细胞细胞因子的分泌格局,表明VIP在抗感染免疫中具有一定的免疫调节作用。Objective To investigate the effects of vasoactive intestinal peptide (VIP)on secretions of IL-1, IL-6, and TGF-β by leydig cell. Methods The testis of the SD rat was digested by collagenase (type Ⅱ), and then filtrated and isolated by Percoll for obtaining high purity leydig cells. After treated by VIP of different dosages, the leydig cells were infected with UU. The secretions of IL-1, IL-6, and TGF-β by the treated leydig cells were detected by MTT, ELISA, and ELISA, respectively. Results VIP increased the secretion of IL-1 but inhibited the secretion of IL-6. The secretion of TGF-β by leydig cells was increased by low-dosage VIP and inhibited by high-dosage VIP. Conclusion When leydig cells are infected by UU, VIP could change the pattern of the secreting of IL-1, IL-6, and TGF-β by leydig cells, which shows that VIP play an definite immunoregulative role in anti-infective immune.
