六神丸含药血清诱导HL-60细胞凋亡实验研究

摘　　要：目的:观察六神丸含药血清诱导白血病细胞凋亡,探讨六神丸治疗急性白血病的部分机制。方法:制备含药血清,用MTT法、胎盘蓝染色法观察六神丸对HL-60细胞的增殖抑制作用。用吉姆萨染色法观察细胞凋亡的形态学变化,流式细胞仪、原位末端标记法(TUNEL)定量检测细胞凋亡程度。结果:六神丸含药血清对白血病细胞有明显的抑制,且随药物血清浓度和培养时间的延长而增加,吉姆萨染色、流式细胞仪检测、原位末端标记检测均证明六神丸含药血清能诱导HL-60白血病细胞凋亡。结论:六神丸含药血清能诱导细胞凋亡,这可能是六神丸治疗白血病的机制之一。Objective：To observe serum contained Liushen pill inducing white blood cell apoptosis and to explore the partial mechanism of Liushen pill in treating acute leukemia. Methods：To produce serum contained Liushen pill. Trypan blue assay and MTT were used to detect the growth inhibition effect of Liushen pill. Gimsa dying assay observes morphology changes of apoptosis. In situ efld-labeling method （POD） and fluorescene-activated cell sorting quantificationally probes apoptosis phenomena. Results：The Suppressive rate of serum contained Liushen pill on HL-60 cell raises with adding dose of medicine serum and prolonging of cultivating time. Gimsa dying assay, in situ end-labeling method （POD） and fluorescene-activated cell sorting all exhibits： HL-60 cells dealed with serum contained Liaashen pill present typical apoptosis morphology. Conclusions： serum contained Liushen pill can induce apoptosis of HL-60 cell. It is one of mechanisms in treating leukemia bv Liushen oill.
