摘 要:目的探讨地塞米松对哮喘豚鼠肺内、C7~T5脊神经节及对应节段脊髓背角内蛋白激酶C(PKC)表达的抑制作用。方法利用免疫组织化学和Western blotting方法,研究哮喘豚鼠肺内、C7~T5脊神经节及对应节段脊髓背角内PKC的免疫反应变化。结果哮喘组豚鼠肺内、C^T5脊神经节及对应节段脊髓背角内PKC平均光密度值明显高于对照组(P<0.01),而地塞米松组则明显低于哮喘组(P<0.01)。结论PKC可能参与哮喘的发病过程,地塞米松抑制哮喘豚鼠PKC的表达可能是其治疗哮喘的作用机制之一。Objective To explore the inhibitory effect of dexamethasone on the expression of protein kinase C(PKC) in the lung, the C7-T5 spinal ganglia and the corresponding spinal dorsal horns of the asthmatic guinea pigs. Methods The alterations of PKC immunoreactivity were investigated by means of immunohistochemistry and Western blot in the lung, the C7-T5 spinal ganglia and the corresponding spinal dorsal horns of the asthmatic guinea pigs. Results The average optic density of PKC in the lung, the C7-T5 spinal ganglia and the corresponding spinal dorsal horns of the asthmatic guinea pigs was much higher than that of the control group( P 〈 0.001 ) and the average optic density of PKC of the dexamethasone group was much lower than that of the asthmatic group( P 〈 0.01 ). Conclusion The present results indicate that the over expression of PKC might be involved in the pathogenesis of the bronchial asthma, and inhibition of the expression of the PKC by dexamethasone may be one of the mechanisms in the treatment of asthma.
