白念珠菌分泌性蛋白酶活力与其对氟康唑耐药性之间关系的研究

摘  要:目的研究白念珠菌分泌性蛋白酶(Sap)活力与其对氟康唑耐药性之间的关系。方法将对氟康唑敏感的白念珠菌培养在含氟康唑浓度逐渐增加的液体培养基中,体外诱导为氟康唑耐药株;运用牛血清白蛋白培养基(BSA)的方法测定了对氟康唑敏感的白念珠菌和对氟康唑耐药的白念珠菌的分泌性蛋白酶活力。结果40株对氟康唑敏感的白念珠菌均在体外被成功的诱导成为对氟康唑耐药的耐药株。耐药株的分泌性蛋白酶活力明显强于敏感株(P<0.001)。结论白念珠菌Sap活力的增加与其对氟康唑耐药性增加是一致的。[Objective] To study the relationship between production of Secreted Aspartyl Proteinase(Sap) and development of fluconazole resistance in Candida albieans. [Methods] Flueonazole-suseeptible Candida albieans isolates were cultured in liquid medium containing increasing FLC concentration from 8.0 to 128 μg/mL, We used the medium containing bovine serum albumin (BSA) to assay the secretory proteinase activity of flueonazole-suseeptible Candida albieans isolate and fluconazole-resistant Candida albicans isolate. [Results] Fluconazole-susceptible Candida albieans isolates following growth in increasing concentrations of flueonazole can be induced into flueonazol-resistant Candida albieans isolate. The Secreted Aspartyl Proteinase activity of fluconazoleresistant Candida albieans strains are higher than fluconazole-suseeptible Candida albieans isolates(P 〈0.001 ). [Conclusion] Enhanced Sap production of Candida albieans isolates corresponded to the development of increased FLU resistance.
