半夏泻心方配伍与诱导BGC-823细胞凋亡关系的血清药理学研究

摘　　要：目的:研究半夏泻心方不同配伍药组含药血清对BGC-823细胞凋亡的影响及其机制。方法:采用MTT法观察半夏泻心方配伍药组含药血清对BGC-823细胞增殖的影响,采用DNA凝胶电泳检测细胞凋亡、S-P免疫组化法检测凋亡相关基因bcl-2的表达。结果:半夏泻心方配伍中辛开组、苦降组和由其组成的辛苦组可明显抑制BGC-823细胞的生长,显著优于辛甘组、苦甘组、甘补组和全方组,呈剂量-效应关系,其含药血清浓度在10%作用48小时后,DNA琼脂糖凝胶电泳可见较典型的“梯状带”,可使bcl-2基因的表达下调。结论:半夏泻心方中辛开、苦降的配伍形式具有抑制BGC-823细胞增殖,诱导细胞凋亡作用,辛开、苦降配伍后(辛苦)有显著的协同增效趋势,而其他配伍药群可能存在拮抗作用。提示辛开苦降法可能是中医药防治胃癌的主要治则治法之一。To study the effects of medicine-containing serum for different combinations of Banxia Xiexin Recipe on BGC-823 cell apoptosis and its mechanisms. Methods： MTT method was used to observe effects of medicine-containing serum for different combinations of Banxia Xiexin Recipe on proliferation of BGC-823 cells,DNA gel electrophoresis used to detect cellular apoptosis, and S-P immunohistochemical method used to investigate expression of apoptosis-related gene bcl-2. Results：Growth of BGC-823 cells was significantly inhibited in the pungent-opening group, the bitter-descending group and the pungent-bitter group, which was significantly higher than that in the pungent-sweet group,bitter-sweet group,the sweet-invigorating group and the complete recipe group（P〈0. 05）,with a dose-effect relation. After action of 10% medicine-containing serum for 48 hours,a more typical ＂scalariform band＂ could be seen on the DNA agar gel electrophoresis, and bcl-2 gene expression was down-regulated, Conclusion：The combination of pungent-opening or bitter descending components in the Banxia Xiexin Recipe has the actions of inhibiting proliferation of BGC-823 cells and inducing cell apoptosis,and after the combination of pungent-opening or bitterdescending components （Pungent-bitter） has a significant tendency to cooperatively increase the effect, but other combinations maybe have antagonism. It is suggested that pungent-opening and bitter-descending method is possibly one of main therapeutic principles and methods for prevention and treatment of gastric cancer with TCM.
