摘 要:目的:观察红花提取物预处理对心肌缺血再灌注损伤的影响。方法:采用离体心脏Langendorff装置,观测经红花提取物预处理后心功能指标和冠脉流量的改变;同时采用在体心脏结扎左冠状动脉前降支制备心肌缺血再灌注模型,在心肌缺血前各组心脏分别用红花提取物或维拉帕米治疗,并贯穿于实验全过程中。缺血再灌注组实施20分钟缺血和40分钟再灌注。分别检测心肌组织中的丙二醛(MDA)含量、超氧化物歧化酶(SOD)和心肌激酶(CK)活性、心率等变化。结果:红花提取物能减少缺血再灌注心肌中的MDA含量,降低SOD和CK活性,减慢心率,改善心功能指标。结论:红花提取物能改善心功能,保护缺血心肌,其机制可能与其清除自由基,抑制自由基的释放有关。To observe effects of pretreatment of extracts of Honghua (Flos Carthami) on ischemia-reperfusion myocardial injury. Methods:Myocardial ischemia-reperfusion injury was induced by ligation of the anterior descending branch of the left coronary artery. The heart was perfused with the extracts of Honghua or verapamil before iscbemia untill the end of the experiment. The ischemia-reperfusion group experienced 20-min ischemia and 40-min reperfusion. Malondialdehyde (MDA) content, superoxide dismutase (SOD) and creatine kinase(CK) activities in the rat myocardial tissue were determined,and the heart rate and cardiac function index were observed. Results :The extract could reduce MDA content ,SOD and CK activities in the ischemia-reperfusion myocardium,decrease the heart rate,and improve the blood pressure,left ventricular systolic pressure (LVSP),and the maximum rates of contraction and relaxation (=dp/dtmax). Conclusion :The extract of Honghua can improve the cardiac function ,and the mechanism may be related with clearence of free radicals and inhibition of release of free radicals in the ischemic myocardium.
