摘  要:目的:观察百合科黄精属中草药玉竹经水醇法提取而来的玉竹提取物A对链脲佐菌素诱导的1型糖尿病小鼠血糖、T淋巴细胞亚群及外周血细胞因子的影响。方法:①实验于2004-05/2004-12在锦州医学院医学实验中心完成。选用C57BL小鼠60只。随机抽取9只作为正常对照组小鼠(只注射等体积枸橼酸钠缓冲液),其余小鼠应用4g/L链脲佐菌素溶液,按40mg/kg剂量腹腔注射制备1型糖尿病模型,1次/d,连续5d,以血糖≥11.4mmol/L,尿糖以上2周,为造模成功,低于此值弃去,造模成功36只。将其随机分为4组,每组9只:模型组,玉竹提取物A1,2,4g/kg组。②正常对照组与模型组腹腔注射生理盐水0.5mL,1次/d,共4周;玉竹提取物A1,2,4g/kg组,分别按1,2,4g/kg剂量腹腔注射0.5mL玉竹提取物A(由锦州医学院免疫教研室潘兴瑜教授提供,玉竹的干燥根茎经水醇法提取而得),1次/d,共4周。③用药4周监测血糖变化,应用流式细胞仪检测用药后1型糖尿病小鼠脾T淋巴细胞亚群含量变化;采用酶联免疫分析法检测用药后1型糖尿病小鼠血清白介素4、白介素10、干扰素γ水平变化。④组间比较采用方差分析。结果:造模失败15只,正常小鼠9只及造模成功小鼠36只进入结果分析。①空腹血糖水平:用药前各组差异不明显(P>0.05),用药后,玉竹提取物各剂量组明显低于模型组(P<0.05~0.01)。②CD4+T淋巴细胞含量和CD4+与CD8+T淋巴细胞比值:模型组明显高于正常对照组和玉竹提取物各剂量组(P<0.05~0.01),CD8+T淋巴细胞含量:模型组明显低于正常对照组和玉竹提取物各剂量组(P<0.05~0.01)。③血清白细胞介素4和10水平:模型组明显低于正常对照组和玉竹提取物各剂量组(P<0.05~0.01),血清干扰素γ水平:模型组明显高于正常对照组和玉竹提取物各剂量组(P<0.05~0.01)。结论:玉竹提取物A经腹腔注射能明显降低1型糖尿病小鼠血糖,其机制与恢复CD4+和CD8+T淋巴AIM: To observe the effect of extraction A from polygonatum odoratum by water alcohol method on blood glucose, T lymphocyte subsets and peripheral blood cells (PBC) induced by cytokine of stroptozotocin (STZ)in mice with typel diabetes, METHODS: ① The experiment was conducted in the Medical Experimental Center Of Jinzhou Medical College from May to December 2004. Sixty C57BL mice were selected, 9 of them were randomly selected as normal control group (only injected of buffer solution of natrium citricum of the same volume), rest mice received iiatraperitoneal injection of 4 g/L solution of streptozotocin at 40 mg/kg and made into type 1 diabetic models once a day for 5 continuous days. Models of blood glucose 11.4 mmol/L and urine glucose above "+" for 2 weeks were considered as success, and those below this value were abandoned. There were 36 successful models, which were randomized into 4 groups with 9 mice in each group: Model group and extraction A 1, 2, 4 g/kg group.② Mice in the normal control group and model group received intraperitoneal injection of 0.5 mL normal saline once a day for 4 weeks; Mice in the extraction A 1, 2, 4 g/kg group received intraperitoneal injection of 0.5 mL extraction A from polygonatum odoratum (obtained from drying rhizome of polygonatum odoratum with water alcohol method and supplied by professor Pan from Staff Room of Immunology, Jinzhou Medical College) at the doses of 1,2,4 g/kg respectively once a day for 4 weeks. ③Changes of blood glucose 4 weeks after administration were monitored, and changes of content of T lymphocyte subsets in spleen of mice with type 1 diabetes after administration were assayed with flow cytometry; Changes of levels of serum interleukin-4 (IL-4), interleukin-10 (IL-10) and interferon 7 in mice with type 1 diabetes after administration were tested with enzyme-linked immunosorbent assay (ELISA). ④ Differences among groups were compared with analysis of variance.
