摘 要:目的:观察电针对脑纹状体缺血性神经元损伤细胞凋亡形态学及基因表达的影响,探索电针对其抗氧应激保护作用可能的分子生物学机制。方法:实验于2003/2004在南京中医药大学针药实验室进行。纳入雄性健康10~12月龄清洁级SD大鼠47只,随机分为6组,即假手术组8只,模型组8只,电针Ⅰ组8只,电针Ⅱ组7只,药物组8只,针药组8只。采用LONGA氏血管线栓法改良,制成脑纹状体缺血性神经元细胞损伤模型。①假手术组:仅作手术血管分离。②模型组:仅右脑动脉丝线栓塞60min。③电针Ⅰ组:造模后,分别电针“百会”、“大椎”穴位。④电针Ⅱ组:造模后,分别电针“百会”、“人中”穴位。⑤药物组:制模手术前30min,用褪黑素溶解于950mL/L乙醇,再以生理盐水配制,按3.2mg/kg腹腔注射。⑥针药组:处理同电针组及药物组。电针治疗中,使用30号0.5寸毫针,接上海G6805电针仪,连续波、频率3Hz,强度2~4mA,时间30min;以针柄轻微颤动、大鼠不嘶叫为宜。全部模型经处理、再灌注24h后处死。各组模型脑纹状体片(包括尾状核、豆状核等)相同部位,连续切取厚5μm组织片5套,分别做苏木精-伊红染色、神经元尼氏体染色、形态学分析、Bax及Bcl-2基因蛋白、Tunnel细胞凋亡数等项目研究,并用各组均值分析、比较。结果:大鼠47只进入实验分析。①电针“百会”、“大椎”穴位30h后形态学观察,梗死灶明显缩小;各病变轻重形态表现较一致,水肿显著减低、纹状体缺血神经元损伤形态学改善显著。②与模型组比较,电针Ⅰ组,电针Ⅱ组,褪黑素组,电针Ⅰ+褪黑素组等治疗组大鼠脑纹状体缺血神经元阳性凋亡细胞数均显著性降低犤(7.93±1.97),(12.8±1.44),(9.46±1.69),(11.78±1.44),(15.94±1.81)个/0.5mm×0.5mm,P<0.001~0.01犦;电针Ⅰ组及褪黑素药物组作用相当,效果显著优于电针Ⅱ组及电针Ⅰ+褪黑素组(P<0.001~0.05)。③电针�AIM: To observe the effect of electroacupuncture (EA) on the neuronal injury and apoptosis morphology of striate body in rats with cerebral ischemia, and explore the possible molecular biological mechanism of EA in the protection of anti-oxidative stress. METHODS: The experiment was performed at Laboratory of Acupuncture Combined with Medication, Nanjing University of Traditional Chinese Medicine between 2003 and 2004. A total of 47 health male SD rats of cleaning grade, aged 10-12 months, were randomized into sham-operation group (n=8), model group (n=8), EA Ⅰ group (n=8),EA Ⅱ group (n=7), medication group(n=8), EA plus medication group(n=8).LONGA modified thread embolism method of blood vessel was applied to establish rat models of ischemic neuronal injury in striate body.①Sham-operation group: Only blood vessel was separated in operation. ②Model group: Only fight middle cerebral arteries occlusion (MCAO) by silk was performed for 60 minutes.③EA Ⅰ group: Alter the establishment of model, EA was applied to the acupoints of Dazhui (DU14) and Baihui (DU20), respectively.④EA Ⅱ group: After the establishment of model, EA was applied to Baihui (DU20) and Renzhong (DU26), respectively, ⑤ Medication group: Intraperitoneal injection of 3.2 mg/kg melatonin, which dissolved in 950 mL/L ethanol and then prepared with saline, was given at 30 minutes before the model establishment. ⑥EA plus medication group: With the same treatment as EA group and medication group, all the models were treated with No. 30 needle(0.5 cun) connected with Shanghai EA apparatus G6805, in the parameters of continuous wave, 3 Hz in frequency, 2-4 mA in intensity for 30 minutes. It was proper that the rats did not hiss and needle handle shivered slightly. Being killed at 24 hours after intervention and reperfusion, all models were cut successively in the same location of striate body (including caudate nucleus, lentiform nucleus, etc)to obtain 5 sets of tissu
