摘 要:目的探讨孕酮对大鼠局部脑缺血再灌注损伤的神经保护作用。方法以栓线法制备大脑中动脉梗阻脑缺血再灌注大鼠模型,利用TTC和免疫组织化学染色法分别观察缺血面积大小和c-fos在大脑皮质的表达。结果①缺血再灌注组脑缺血面积明显大于缺血组(P<0.05);各实验组不同时间给予孕酮后脑梗死面积明显小于相应对照组(P<0.05)。②缺血组和缺血再灌注2h组可见Fos阳性细胞主要分布于缺血区周围的半影区;各实验组不同时间给药后扣带回皮质Fos阳性细胞数均高于相应对照组(P<0.05)。结论孕酮对大鼠局部脑缺血和再灌注具有明显的神经保护作用。Objective The neuroprotective effects of progesterone was evaluated in rats with middle cerebral artery occlusion (MCAO) and reperfusion. Methods The rats model of MCAO and reperfusion was produced with a string that was inserting into the internal carotid artery to occlude MCA. TTC staining and immunocytochemistry were used to analyze infarction size and c-fos expression in region arounding infarction area. Results ①The infarction size in rats with MCAO and reperfusion was significantly larger than that in rats with MCAO without reperfusion( P 〈0.05). The infarction size in rats with MCAO pretreated with progesterone were all smaller than that in the ischemic rats in corresponding time points. ②c-fos expression was mainly distributed in ischemic penumbra with MCAO combined with or without reperfusion. The numbers of Fos-positive neurons with pretreatment of progesterone were larger than those in the ischemic rats significantly( P〈0.05). Conclusion The results may suggest that progesterone has some neuroprotective effects to MCAO rats whether combined with reperfusion or not.
