摘 要:以骨形态汁量学方法探讨不同剂量、周期性腹腔注射羟乙膦酸钠对切除双侧卵巢大鼠的骨矿化和骨重塑的影响。结果表明,本实验所用的两种剂量(51mg/kg和36mg/kg)的羟乙膦酸钠使软骨细胞和软骨基质堆积,骺板显著增厚,干骺端类骨质显著增多,骨动力学参数如纵向骨生长率、活性生成表面百分比和骨矿化速率无法检出。说明过量的羟乙膦酸钠使骨软骨基质和骨基质矿化受阻,进而骨吸收被抑制,骨转换率降低,骨重塑发生障碍。这是临床应用羟乙膦酸钠防治骨质疏松症过程中应该引起注意的。Fifty-four female Wistar rats,with an average weight of 266g and bilaterally ovariectomized, were used to study the effects of different doses of etidronate disodium(51mg/kg, 36mg/kg and. 25mg/kg BW) on bone mineralization and remodeling. HEBP was given intraperitoneally for 5 days and followed by a resting period of 25 days. This cycle was repeated 3 times and up to 90 days totally. Serum Ca,P contents and AKPase activity,bone Ca and P contents were determined. Bone histomorphometry was performed on undecalcified sections of the proximal tibia by general and fluorescent microscopy. Data showed that the higher doses HEBP (51mg/kg and 36mg/kg) induced marked accumulation of chondrocytes and chondromatrix in epiphyseal plate and significant increase of osteoid in metaphysis,serum AKPase activity and bone Ca content were significantly reduced,and the bone kinetic parameters could not be revealed. The results suggested that the overdose HEBP by intraperltoneal injection resulted in the excessive suppression of mineralization and then proceeding to the inhibition of bone resorption and impeding bone remodeling. Lower dose HEBP (25mg/kg) presented a little therapeutic effects but its osteoid also increased though lower than that of the higher doses.
