摘 要:目的探讨三苯氧胺(TAM)对小鼠精子的毒性作用。方法将雄性小鼠随机分成阴性对照组、TAM(低、中、高剂量)组和阳性对照组,分别以生理盐水、TAM[0.02、0.22、mg/(kg.d)]和环磷酰胺[40 mg/(kg.d)]连续灌胃20 d,于末次灌胃后15 d处死小鼠,称取睾丸、附睾重量,计算精子活动度、精子数与畸形率。结果①高、中剂量TAM组的精子活力与精子数量显著低于阴性对照组(P<0.01);②高、中剂量TAM组的精子畸形率高于阴性对照组,差异有极显著性意义(P<0.01);③高剂量TAM组的睾丸指数小于阴性对照组,差异有显著性意义(P<0.05)。结论TAM对小鼠的精子有毒性作用。Objective To explore the toxicity of tamoxifen (TAM) at different doses on sperm in male mice. Methods The male mice were randomly divided into negative control group, TAM (low dose, middle dose and high dose) groups and cy toxan group, receiving normal saline, TAM [0.02, 0.2, 2 mg/(kg · d)] and cytoxan [40 mg/(kg · d)] respectively by gavage once a day for successive 20 days. The mice were sacrificed on the 35th day after the first gavage. The weight of testicle and epididymis, the quantity, activity, morphology of sperms were examined. Results Compared with those in the negative control group, the spermatozoa motility and count in the high and middle dose TAM groups were significantly reduced (P〈0.01). Sperm deformation ratio was significantly higher in high and middle dose TAM groups than in the control groups (P〈0.01). The index of testes in the high dose TAM group was obviously lower than in negative control group (P〈0.05). Conclusion TAM has toxicity on sperm in male mice.
