雷公藤单体T_4对类风湿关节炎患者周围血单个核细胞及滑膜细胞产生肿瘤坏死因子的影响  被引量:6

The Effects of a Single Active Ingredient(T4)of Tripterygium Wilfordii Hook on the Production of Tumor Necrosis Factor by the Peripheral Blood Mononuclear Cells and Syuovium Cells of Rheumatoid Arthritis Patients

摘  要:观察了经雷公藤单体T_4处理2h后对正常人和类风湿关节炎(RA)患者外周血单个核细胞(PBMC)及RA患者消化的滑膜单个细胞(RA-DSSC)分泌TNF的影响。在35ng/ml浓度下,正常人PBMC(n=10)及RA-PBMC(n=6)T_4处理组产生TNF活性水平明显低于对照组(P<0.001)。延长T_4作用时间至6h,RA-PBMCT_4处理组TNF活性水平下降更为明显(P<0.001),表明T_4可抑制PBMC体外产生TNF活性水平,其对RA-PBMC作用不同时间的抑制率分别为25.54%及40.51%(P<0.01),说明T_4抑制作用可能与作用时间有关。同时T_4亦可抑制RA-DSSC体外产生TNF活性水平(P<0.01),提示T_4有望成为一新的植物性治疗RA药物。Encouraged by good therapeutic results of the semipurmd preparations of TripterygiumWilfordii Hook(TWH)(code name T Ⅱ)a single active ingredient of TWH with the codename of T4 was obtained.The in vitro effects of T4 on the bioactivity of TNF produced byPBMC of normal subjects(n= 10)and RA patients(n=6)or the RA digested synovium sin-gle cells(DSSC)were studied.The results showed that under pretreatment of PBMC ofhealthy persons with T4(35ng/ml) for two hours,TNF were reduced to 36.13±1.75%incomparison with the control of 63.33 ± 2.51%(P<0.001),with an inhibition rate of40.31%.Similarly,the inhibition rate for the PBMC of RA patients(P<0.001),25.54%.The fact that coculture of PBMC and T4 for 6 hours instead of treatment with T4for 2 hours caused an inhibition rate of 40.51%(P<0.01),might indicate that time of in-teraction was also a factor of importance.T4 also reduced TNF secretion by RA-DSSC from72.1±6.1%to 51.6±2.0%with an inhibition rate of 28.4%.The aformentioned indicatesthat T4 appears to possess suppressive actions on production of TNF by PBMC and DSSC ofRA patients.Therefore,it is possible that T4 may be used clinically for RA patients.
