摘 要:采用花粉管通道法,将青菜株系76F5、14号大白菜、紫阳等三个品种的总DNA及质粒pActl-DDNA导入青菜605品系,收获青菜种子.种子发芽后分别提取约两周苗龄的小苗总DNA,并经EcoRI酶切,以gus基因片段为探针做分子杂交.证实了外源gus基因已整合到青菜605品系中;田间观察还获得了一株与供体大白菜花形相似的变异株.otal DNAs of Brassica chinensis L. strain 76F5, Brassica pekinensis L.,Brassica oleracea var rubra L. and plasmid pActl-D were introduced into Brassica chinensis L. variety 605 via pollen-tube pathway. Seeds from the foreign DNA-treated plants were harvested and allowed to germinate. Hybridization of the gus gene fragment probe to products of EcoR I-digested total DNAs from various plants were carried out and hybridization bands were detected in the total DNAs of progenies from foreign DNA-treated plants. A variant similar to the donor Brassica pekinensis L. was obtained among progenies.
