摘  要:目的克隆日本血吸虫(Sj)FKBP12基因全长cDNA,并进行免疫组织定位。方法从成虫RNA中RT-PCR扩增SjFKBP12基因完整的编码阅读框后,将其分别克隆入pGEX-4T-1和pEGFP-N1载体中;DNA免疫后制备抗血清,间接免疫荧光法对FKBP12进行组织定位。结果将SjFKBP12基因成功克隆入pGEX-4T-1和pEGFP-N1载体中;FKBP12定位于Sj成虫的被膜上。结论成功克隆了SjFKBP12基因,并对FKBP12蛋白在Sj成虫中进行组织定位,为将其进行疫苗等研究打下了基础。The cDNA of the FKBP12 gene of Schistosoma japonicum ( Sj ) was cloned and its immunohisto-loacalization was investigated in the present study, in which the full length of this gene was obtained from the adult worm RNA of S. japonicum by RT-PCR, and then was cloned into vectors of pGEX-4T-1 and pEGFP-N1, and indirect immunofluorescence assay was used to find its histo-localization in various tissues. It was demonstrated that the FKBP12 gene eDNA sequence could be cloned successfully into vectors pGEX-4T-1 and pEGFP-N1, and localized on the tugment of S. japonicum. The result of this investigation provides basis for the further studies on vaccine development.
