摘 要:目的观察冲击波负压(BUP)暴露后早期豚鼠耳蜗毛细胞胞核的病理形态学变化。方法将豚鼠在负压峰值为-44.5^-48.8kPa的中等强度BUP暴露3次,于BUP暴露后7d解剖取出耳蜗,应用DNA荧光染料Hoechst 33342显示毛细胞核,在荧光显微镜下毛细胞病理损害情况并进行计数。结果BUP暴露后7d,豚鼠耳蜗毛细胞可见胞核肿胀、胞核缺失、胞核固缩3种病变,以核肿胀发生最高,其次是核缺失,核固缩最为少见,3种病变的总发生率达(17.3±3.2)%,明显高于正常对照组(P<0.001)。结论BUP暴露可导致耳蜗外毛细胞核明显的病理性改变,这种改变可能是造成听力损失的主要原因之一。Objective To observe the morphologic changes in the cochlear hair cell nuclei of the guinea pigs following exposed to blast underpressure (BUP). Methods The guinea pigs were exposed to the moderate BUP at peak underpressure between -44.5 kPa and -48.8kPa for three times. The cochleas were collected for the examination of the hair cell nuclei 7d after the BUP exposure. Hoechst 33342, a fluorescent DNA stain, was used to identity the nuclei and the specimens were examined under a fluorescence microscope for quantitative assessment of the hair cell nuclear damage. Results There were three types of morphological changes in the damaged outer hair cell nuclei of the guinea pigs after BUP exposure, nuclear swelling, nuclear condensation and nuclear missing. Nuclear swelling was more frequent than nuclear missing and nuclear condensation. The total incidence rate of these damages was (17.3±3.2)% ,it was obviously more than that of the control group (P〈0. 001 ). Conclusion The results suggest that exposure to BUP can lead to visible pathlological changes of the cochlear out hair cell nuclei that may be one of main causes of hearing loss.
