摘  要:目的探讨bcl-2蛋白家族与庆大霉素诱导耳蜗螺旋神经节细胞凋亡的关系。方法对豚鼠进行庆大霉素连续肌肉注射,150mg/kg×10d,分别于停药后1、3、7、14d各处死豚鼠8只,取耳蜗,做平行蜗轴的石蜡切片,用免疫组织化学方法检测连续用药后不同时间耳蜗螺旋神经节细胞bcl-2及Bax的表达变化情况。结果耳蜗螺旋神经节细胞bcl-2表达增强,3d时表达达到高峰,7d时有所下降,14d时bcl-2表达明显减弱;Bax的表达在3d时达到高峰,7d时表达较3d时下降,14d时Bax表达减弱。结论庆大霉素作用下,bcl-2和Bax表达发生变化,bcl-2蛋白家族参与了庆大霉素诱导的耳蜗螺旋神经节细胞凋亡。Objective To investigate the expression of bcl-2 and Bax proteins, regulators of apoptosis in guinea pig cochlear injury by gentamicin. Methods Gentamicin was administred 150mg/kg × 10d on guinea pig consecutively. The expressions of bcl-2 and Bax proteins were examined by immunohistochemical method in cochlea of normal and gentamicin-treated animals. Results bcl- 2 and Bax were expressed in the cochlea of normal guinea pig in the low level. After gentamicin applied to the animals(GM group), the expression of the two factors increased to the peak in 3d,and then decreased. The descent tendency of the bcl-2 expression was more obvious than that of Bax. The expression level of bcl-2 had a tendency to be lower than control in 14d after gentamicin administrated, while that of Bax was higher than control(P〈0.05). Relatively speaking, expression of Bax increased, and expression of bcl-2 decreased. Apoptosis of inner ear cells was promoted. Conclusion The expressions of bcl-2 and Bax protein have changes under gentamicin administration, bcl-2 family proteins should play an important role in regulation of cochlear cells apoptosis.
