摘  要:目的检测慢性脑缺血模型大鼠学习记忆能力的改变、海马神经元的损伤和脑内过氧化水平的改变,并观察何首乌成分四羟基二苯乙烯苷(2,3,5,4-′tetrahydroxystilbene-2-O--βD-glucoside,TSG)在上述各方面是否具有显著的改善和保护作用。方法雄性SD大鼠双侧颈总动脉永久性结扎制备慢性脑缺血致痴呆模型。采用Morris水迷宫和避暗实验检测大鼠的学习记忆能力,用尼氏染色方法检测海马神经元细胞,采用生化方法检测皮层及海马脂质过氧化物丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-PX)活性。结果慢性脑缺血模型组大鼠空间学习记忆能力和被动回避学习记忆能力较假手术对照组明显降低,海马神经元细胞减少,尼氏体显著减少,皮层和海马MDA含量较假手术对照组明显升高,GSH-PX活性显著降低。TSG灌胃给药可显著改善模型动物的空间和被动回避学习记忆能力,减轻脑缺血后海马神经元的损伤,增强皮层和海马GSH-PX活性,降低脑组织MDA含量。结论TSG可改善双侧颈总动脉结扎致痴呆模型大鼠的学习记忆能力,减轻海马神经元的损伤,提高脑内抗氧化能力,降低其过氧化水平。OBJECTIVE To detect the changes in learning and memory ability, the injury of hippocampal neurons and the intraccphabc peroxidation level in model rats with chronic cerebral ischemia, and to investigate whether 2,3,5,4′-tetrahydmxystilberte-2-O-β-D-glucoside (TSG), the main effective component of polygonum multiflorom, has significant improving and protecting action in the above aspects.METHODS The chronic cerebral ischemia models wcrc induced by the 8-week permanent ligation of bilateral carotid arteries in malc SD rats. Morris water mazc and the step-through test were used to detect the learning and memory ability. Nissl's staining was used to show the status of hippocampal neurons. The biochemical methods were used to examine the MDA content and GSH-PX activity in cerebral cortex and hippocampus.RESULTS In model rats, the spatial learning and memory ability,passivc avoidance learning and memory ability were severely impaired, hippocampal neurons and Nissl's bodies wcre notably reduced, GSH-PX activity significantly decreased, and MDA level increased. The administration of TSG markedly improved the learning and memory ability of model rats, mitigated the injury of hippocampal neurons, increased the GSH-PX activity and decreased the MDA level in both cortex and hippocampus. CONCLUSION TSG can improve the learning and memory ability of chronic cerebral ischemic model rats, mitigate thc injury of hippocampal neurons, increase the antioxidation ability and decrease the peroxidation level in cerebral cortex and hippocampus.
