摘  要:作者通过不同浓度的低密度脂蛋白(LDL)与培养的新生小牛腔静脉内细胞(EC)孵育后,应用凝血酶、蛋白C和S-2238的发色底物显色法,检测单层EC膜上血栓调节蛋白(TM)辅助激活产生活化蛋白C(APC)的量,以此观察TM功能的变化。结果显示:LDL孵育浓度≥25μg/ml后,APC生成受抑;两者间呈负相关(r=-0.87,n=286,P<0.001);随着LDL孵育时间延长,TM受抑加重,6h孵育后受抑较2h为著;动力学分析表明LDL预处理使APC整个生成过程受抑;降低了APC生成率,早期尤甚,使生成率-反应时间曲线的曲率半径增大;APC生成反应速率出现前期减慢、后期加速的变化等。这些变化提示高脂可以通过抑制血管壁TM功能来影响机体局部抗凝功能。The changes of thrombomodulin (TM) activity were observed after cultured newborn bovine caval endothelial cells (EC) were incubated with variable concentrations of low density lipoprotein (LDL)in different incubation time by measurement of TM-dependent protein C activation on the surface of EC monolayers with thrombin and purified protein C,and S-2238.The results showed that after incubation with more than 25μg/ml LDL,activated protein C(APC)generation was significantly decreased,indicating that TM activity had been suppressed.There was a considerable negative correlation between LDL concentration and TM activity(r=-0.87,n=286, P< 0.001).The longer LDL incubation time,the more inhibition of TM activity achieved.The inhibition of six hours incubation was significantly more than that of two hours.Dynamic analysis showed that pre-incubation with LDL resulted in inhibition of whole process of APC generation,and decline of APC generation rate,especially in early period,so the radius of APC generation rate and reaction time curve was enlarged.The reaction rate of APC generation decreased in early stage but increased later.These data suggest that elevated blood level of LDL may contribute to decreasing the function of local anticoagulation by inhibiting the activity of TM on the vessel well.
