摘　　要：目的探讨佐剂FQ_2与日本血吸虫DNA26Kda疫苗共同作用下小鼠的免疫状态。方法选取4～6周龄BALB／C小鼠52只,分成生理盐水组、FQ2组(A)、Sj26GST组(B)和FQ_2+SJ26GST组(C)等4组,于第0、2、6周各免疫1次,接种后4周以日本血吸虫尾蚴攻击感染,感染后6W剖杀小鼠,分析脾脏T淋巴细胞CD4^+和CD8^+细胞率,测定血清中特异性IgG水平。结果 B组和C组均可诱导CD8^+淋巴细胞的增殖,C组升高更为显著,CD4^+／CD8^+比率倒置,也均可于4周后诱导小鼠产生高滴度的IgG抗体,二组间差异无显著(P>0．05)。结论 PQ_2对Sj26GST DNA疫苗有明显的增效作用,推测增效作用主要由细胞免疫所介导。Objective To study on the action mechanism of adjuant FQ2 on the enhancement of immunogenicity of Sj26GST DNA vaccine against Schistosoma japonicum infection. Methods BALB/C mice were divided into 4 groups, and injected with FQ2 ＋ Sj26GST DNA vaccine, Sj26GST DNA vaccine, adjuant FQ2 and NS respectively. After 4w of immunization, mice in 4 group were chanllenged with Schistosoma japanica cercariae and killed on the 6th week of infection. Specific IgG antibody in the serum of mice and CD4^＋/CD8^＋ T lymphocytes were also detected by ELISA and flow cytometry method. Results The CD8^＋ T lymphocytes proliferated much more significant in the mice of FQ2 ＋ Sj26GST DNA vaccine group than that of Sj26GST DNA vaccine group, No significant difference in induced titers of specific IgG antibody was observed between the above two vaccine groups. Conclusion The adjuant FQ2 can enhanced the action of Sj26GST DNA vaccine against Schistosomajapanica infection in mice by promoting cellular immune response.
