摘 要:目的:采用离体鼠心急性冬眠模型,探讨心脏去甲肾上腺素在急性心肌冬眠形成中的作用。方法:实验于2004-03/2005-02在华中科技大学同济医学院附属协和医院心血管病实验室完成。选择健康雄性SD大鼠49只,采用大鼠离体心脏急性冬眠模型,随机分成7组,即平衡组、冬眠组、多巴酚丁胺组、对照组、利血平组、对照-酪胺组及利血平-酪胺组,每组7只,每组先随机选6只进行实验,如果6只皆成功,则结束实验,否则进行第7只实验。测定利血平处理及生理盐水处理大鼠冬眠时冠脉流量、压力心率乘积、乳酸脱氢酶漏出量(衡量缺血对心肌细胞膜的损伤,用酶联分光光度分析法检测)、心肌三磷酸腺苷(虫荧光素酶法)、磷酸肌酸(反向离子对液相色谱法)和糖原(化学比色法)的含量,用原位免疫组化法测定心肌中Bcl-2、Bax表达产物(吸光度),用末端探针标记法检测凋亡的心肌细胞(细胞核蓝染即为凋亡细胞),并应用透射电镜观察心肌超微结构。结果:平衡组、冬眠组、对照组及利血平组各有1只大鼠死亡,各组均补充1只大鼠继续实验,进入结果分析42只大鼠,每组6只。①大鼠的心率比较:对照-酪胺组大鼠的心率明显增加(从308.2次/min到409.8次/min,P<0.05),利血平-酪胺组大鼠的心率无明显增加(从275.8次/min到285.7次/min,P>0.05),提示利血平耗竭了心脏交感神经末梢去甲肾上腺素的储备。②鼠心急性冬眠时冠脉流量、压力心率乘积、乳酸脱氢酶漏出、心肌三磷酸腺苷、磷酸肌酸、糖原、Bcl-2,Bax表达产物平均吸光度值和凋亡心肌细胞数比较:利血平处理大鼠分别为1.37mL/min,(2.0±0.5)×103mmHg·次/min,13.83mU/(g·min),1.98μmol/g,2.95μmol/g,4.36mg/g,0.3436,0.2733和2.0个/视野;生理盐水处理大鼠分别为1.08mL/min,(2.3±1.0)×103mmHg·次/min,14.75mU/(g·min),1.53μmol/g,2.33μmol/g,3.37mg/g,0.3880,0.3148和1.5个/视野,两组间差异无显著性意�AIM: To investigate the role of myocardial noradrenaline in the development of acute myocardial hibernation with short-tempered hibernation models of isolated rats. METHODS: The experiment was done in the Cardiovascular Laboratory of Union Hospital Affiliated to Tongji Medical College, Huazhong University of Science and Technology from March 2004 to February 2005. Forty-nine healthy male SD rats were selected and randomly divided into 7 groups with short-tempered hibernation models of isolated rats: balance group, hibernation group, dobutamine group, control group, reserpine group, control-tyramine group and reserpine-tyramine group with 7 rats in each group. Six of 7 rats were experimented on in each group, if all succeeded, the experi- ment would be over, or the seventh was used. The coronary flow, the pressure-rate product, the leakage of lactate dehydrogenase (ischemia on injury of myocardial cell membrane was detected wtih linked-enzyme spectropho-tometric assay), the content of adenosine 5'-triphosphate (biol method), the content of phosphocreatine (reverse phase-high performance liquid chromatography method), the content of glycogen (chemistry colorimetry method), the Bcl-2 products and Bax products(In situ immunohis-tochemistry)(absorbance), the myocyte apoptosis (In situ terminal deoxynucleotidyl mediated dUTP nick end labeling,TUNEL) and the myocardial ultrastrncture(transmission electron microscope) were determined in rats of reserpine group and control disposal when hibernated. RESULTS: One rat failed to finish the experiment in each group of balance group, hibernation group, control group and reserpine group, and the seventh was used. Totally 42 rats were involved in the result analysis with 6 rats in each group.①Comparison of heart rates: The heart rate increased significantly (from 308.2 times per minute to 409.8 times per minute, P 〈 0.05) in the control- tyraminc group, but it increased insignificantly in reserpine-tyramine group (275.8 times
