摘 要:目的明确产生天然抗角蛋白自身抗体(anti-keratin autoantibody,AK auto Ab)的B细胞的类群和定位。方法取SPF级C57BL/6小鼠的腹腔细胞及脾细胞,进行体外培养,采用ELISA方法分析培养细胞分泌AK auto Ab的水平,ELISPOT方法分析分泌AK auto Ab的细胞数。结果腹腔细胞中分泌AK auto Ab的细胞数远多于脾细胞,其分泌的抗体滴度也显著高于脾细胞。结论AK auto Ab主要来源于腹腔B细胞,B1细胞可能是产生AK auto Ab的主要细胞。Objective To clarify the subsets and location of B cells producing natural anti-keratin autoantibody (anti - keratin autoantibody,AK auto Ab). Methods Cells from spleen and peritoneal cavity of C57BL/6 mice were cultured in vitro, and ELISPOT was utilized to detect and calculate the cells that can identify keratin, and ELISA was applied to measure anti-keratin IgM level in culture supernatant. Results Number of Cells producing anti-keratin antibody in peritoneal cavity are greatly higher than those in spleen. And anti-keratin antibody level in the supernatant of cultured peritoneal cell was much higher than that of spleen cells. Conclusion Anti-keraitn autoantibody mainly originate from peritoneal cavity B cells, and B1 cells in the peritoneal cavity maybe the main source of anti-keraitn autoantibody.
