摘  要:目的研究HSP90的表达与乳腺癌细胞侵袭转移能力的相关性。方法采用RTPCR和Westernblot检测HSP90在MDAMB435s和MDAMB231细胞中mRNA水平和蛋白质水平的表达差异。Transwell小室人工重组基底膜侵袭迁移实验检测经HSP90抑制剂Geldanamycin(GA)处理前后两株细胞侵袭迁移能力的改变。结果MDAMB435s细胞中HSP90α的mRNA表达水平明显高于MDAMB231细胞(P<0.01),MDAMB435s和MDAMB231细胞中HSP90β的mRNA表达水平没有明显差异(P>0.05),MDAMB435s细胞中HSP90的蛋白质表达水平明显高于MDAMB231细胞(P<0.01)。两株细胞用药前侵袭迁移能力无明显差异(P>0.05),HSP90的抑制剂GA对MDAMB435s和MDAMB231细胞的侵袭迁移能力均呈现明显抑制作用(P<0.01,P<0.05),对MDAMB435s细胞的抑制作用更明显(P<0.01)。结论HSP90的表达影响乳腺癌细胞侵袭转移能力。Objective To investigate the correlation between the HSP90 expression and invasion and metastasis potential of human breast cancer cell line. Methods The mRNA expression level of HSP90 was detected by RT-PCR. The protein expression level of HSP90 was measured by Western blotting. The effects of HSP90 inhibitor Geldanamycin (GA) on the invasion and motility abilities of human breast cancer line MDA- MB-231 and MDA-MB-435s in vitro were explored by transwell chamber. Results The mRNA expression level of HSP90α in MDA-MB-435s cell was higher than that of MDA-MB-231 cell (P 〈 0.01 ). The mRNA expression level of HSP90β had no difference (P 〉 0.05 ) between the two cell lines. The protein expression level of HSP90 in MDA-MB-435s cell was higher than that of MDA-MB-231 cell (P 〈0.01 ). The invasive and metastatic potential in human breast cancer cell MDA-MB-435 s and M DA-MB-231 had no difference (P 〉 0.05 ). After treated with GA, the invasive and metastatic potential in human breast cancer cell MDA-MB-435s and MDA-MB-231 was suppressed significantly ( P 〈 0.01, P 〈 0.05 ), but the effects of HSP90 inhibitor GA on invasive and metastatic potential of MDA-MB-435s cell was more significant than that of MDA-MB-231 cell (P 〈 0.01 ). Conclusion The abilities of invasion and metastasis of breast cancer cells are related to the expression of HSP90.
