摘  要:目的寻找影响神经元和胶质细胞分化的外在因素,探索胰岛素样生长因子_1(IGF_1)对神经干细胞(NSCs)增殖和分化的影响。方法从E14的SD大鼠大脑中分离神经干细胞,分别用IGF_1、EGF和bFGF处理,台盼蓝染色确定活细胞数量,BrdU标记并分析细胞增殖能力,免疫细胞化学法鉴定神经干细胞、新生细胞和分化细胞。结果分离得到的细胞表现出NSCs特性,用IGF_1、EGF和bFGF处理后均显示增殖效应,并能分化为神经元和胶质细胞样细胞。结论IGF_1能促进NSCs增殖,并诱导其向神经元样和胶质细胞样细胞分化。Objective To observe the effects of insulin-like growth factor-1 on the proliferation and differentiation of neural stem cells (NSCs) and to search the exogenous factors which influence the differentiation of neurons and glia. Methods NSCs were isolated from E14 SD rat brains and then treated with IGF-1, epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) respectively. The cells were dyed with trypanblue to take count of living cells. BrdU was used to lable cells to evaluate the cell proliferation. NSCs, newborn cells and the differentiated cells were identified by immunocytochemistry. Results The cells which were isolated from embryonic rat brains displayed the characteristic of NSCs. NSCs treated with IGF-1, EGF and bGFG showed proliferating capability and differentiated into neuron-like and astrocyte-like cells. Conclusion IGF-1 induce NSCs to proliferate and differentiate into neuron-like and astrocyte-like cells in vitro.
