无症状人芽囊原虫感染调查及PCR鉴定

摘　　要：目的调查当地居民人芽囊原虫感染情况,用PCR鉴定方法进行初步的分子生物学研究。方法收集当地居民粪便标本,碘染色法镜检人芽囊原虫,体外分离培养并观察其形态及繁殖方式,提取人芽囊原虫基因组DNA,用针对人芽囊原虫SSu rDNA的特异性引物进行PCR鉴定。结果共调查415例当地居民,镜检发现4例阳性,均为无症状人芽囊原虫感染者,阳性率0.96%,未见其他寄生虫合并感染。分离培养观察形态主要为空泡型和颗粒型,未发现阿米巴型和包囊型。繁殖方式以二分裂为主,偶见出芽生殖及三分裂,未发现裂体生殖。PCR扩增获得特异性的阳性条带。结论无症状人芽囊原虫感染率较低,可以通过扩增其SSu rDNA进行鉴定。Objective To investigate infection status of blastocystis hominis in local asymptomatic residents, primarily molecular biological study on B. hominis with PCR methods. Methods Fecal specimens were collected from local people, B. hominis were examined under light microscopy by using iodine staining method. Isolated culture of B. hominis was done, morphous and reproductive mode of cultured B. hominis was observed. Genomic DNA were extracted and PCR identification were done with specific primers to SSu rDNA. Results 415 residents were investigated and 4 were positive, all of which were asymptomatic B. hominis infection. Infection rate is 0.96% and other parasites were not found. All of the 4 B. hominis were cultured successfully. Morphous of B. hominis were mainly vacuolar form and granular form, cyst form and ameboid form were not found. Binary fission is the main method of reproduction for B. hominis, gemination and ternary fission were found occasionally, and schizogony was not found. Genomic DNA of B. hominis was extracted and specific PCR product was amplified successfully with specific primers to SSu rDNA. Conclusion Infection rate of asymptomatic B. hominis in local people is low. B. hominis can be identified by PCR amplified with specific primers to SSu rDNA.
