解毒通络保肾胶囊对糖尿病大鼠肾脏超微结构的影响

摘　　要：目的观察中药解毒通络保肾胶囊对糖尿病(DM)大鼠肾脏超微结构的保护作用。方法将实验动物分为正常对照组、DM组、罗格列酮治疗组和解毒通络保肾胶囊治疗组。检测各组第12周的血糖、肾功能、尿白蛋白等变化。光镜、电镜下观测肾内纤维连接蛋白(FN)、Ⅳ型胶原(ColⅣ)和基质金属蛋白酶抑制物-2(TIMP-2)蛋白表达水平。结果解毒通络保肾胶囊治疗组较DM组血糖水平显著降低(P<0.01),尿白蛋白明显减少(P<0.05),尿素氮和血肌酐水平下降,肾脏肥大指标改善(P<0.01);明显降低TIMP-2蛋白表达;FN和ColⅣ蛋白表达亦低于DM组(P<0.01)。结论解毒通络保肾胶囊对DM大鼠肾脏超微结构病变有保护作用,其机制可能与改善糖代谢,减少细胞外基质的积聚有关。Objective: To observe the protective effect of Chinese medicine-jiedutongluobaoshen capsule on renal ultrastructure of diabetic rat. Methods: Rats were divided into four groups: normal group, diabetic model, western medicine and jiedutongluobaoshen capsule groups, the changes of blood sugar, renal function and urinary albumin were determined, the expression of renal FN, COLⅣ and TIMP-1 were observed by light scope and electron microscope.Results:Jiedutongluobaoshen capsule could lower the blood sugar of diabetic rats, significantly decrease urinary albumin, BUN and SCr levels, improve renal hypertrophy index and obviously lower the expression of TIMP-1 protein, decrease the expression of FN and COLⅣ protein, all these were different from diabetic group. Conclusion:Jiedutongluobaoshen capsule could effectively protect the renal ultrastructure of diabetic rat, its mechanism maybe improve glycometabolism and decrease the accumulation of extracellular matrixes.
