摘  要:目的研究大鼠哮喘模型肺组织信号转导子和转录激活因子4(STAT4)蛋白及其mRNA表达的变化,探讨其在哮喘性气道炎症形成中的作用。方法复制大鼠哮喘模型,采用免疫组织化学和组织原位杂交法检测哮喘组及对照组大鼠肺组织STAT4蛋白及其mRNA的表达。结果STAT4蛋白及其mRNA在两组大鼠气道平滑肌细胞和肺细小动脉的血管平滑肌细胞及内皮细胞上均呈不同强度的阳性表达。对照组STAT4蛋白及其mRNA含量均高于哮喘组(均P<0.01)。结论哮喘大鼠气道平滑肌细胞、血管平滑肌细胞及内皮细胞STAT4蛋白及其mR-NA的表达被抑制。Objective To investigate the expression of signal transducer and activator of transcription 4 (STAT4) and its mRNA in lungs of asthmatic rats. Methods The STAT4 and its mRNA expression in lung tissue of asthmatic rat model and the control rats were examined by immunohistochemistry and hybridization in situ methods, respectively. Results The STAT4 and its mRNA was positively expressed in the airway smooth muscle cells, the pulmonary arterial smooth muscle cells and endothelial cells. Compared with control group, STAT4 and its mRNA expression were significantly reduced in asthma group (P〈0.01). Conclusion Expression of STAT4 and its mRNA in lung tissue are significantly depressed in asthmatic rats.
