摘 要:选用66只18周龄商品代伊莎蛋鸡,随机分为3组,每组22只。对照组饲喂基础饲粮,试验1、2组在基础饲粮中分别添加60和180 mg/kg锌(以ZnSO4.H2O形式),10周后屠宰采样。采用相对定量反转录多聚酶链式反应(RT-PCR)方法检测不同锌添加水平对蛋鸡肝脏和肾脏中金属硫蛋白(Metallothionein,MT)mRNA表达水平的影响。结果表明:试验1组蛋鸡肝脏中MT mRNA表达丰度显著高于试验2组和对照组(P<0.05),而试验2组与对照组之间无明显差异(P>0.05)。试验1组蛋鸡肾脏MT mRNA表达丰度显著高于试验2组(P<0.05),而试验1、2组与对照组之间均无显著性差异(P>0.05)。在饲粮中添加低水平锌可显著上调蛋鸡肝脏和肾脏中MTmRNA的表达。The present experiment was conducted to determine the changes of hepatic and renal metallothionein mRNAs expression in response to different levels of zinc supplementation in ISA laying hens . The laying hens were divided into the groups of control, treatment 1 (60 mg/kg ) and treatment 2 (180 mg/kg) at 18 weeks of age and sacrificed after 10 weeks for tissue sam piing. The levels of mRNA expression were quantified with RT-PCR, using β-aetin as an internal standard. The results showed that zinc supplementation at 60 mg/kg, but not 180 mg/kg, signif icantly increased the hepatic MT mRNA level compared with the control (P〈0.05). Additionally, the level of renal MT mRNA expression was significantly higher in 60 mg/kg compared with that in 180 mg/kg group (P〈0. 05), although no significant differences were observed between control and either of the two treatment groups. The results suggest that lower level of zinc supplementation significantly up-regulate MT gene expression in the liver and kidney of the laying hens.
