加减温经汤对寒凝血瘀模型大鼠卵巢功能的影响

摘　　要：目的观察加减温经汤对寒凝血瘀模型大鼠卵巢功能的影响，探讨加减温经汤治疗寒凝血瘀型妇科疾病的作用机制。方法将大鼠置于0～1℃冰水中造成寒凝血瘀大鼠模型，采用阴道涂片检测大鼠动情周期；剖取子宫、卵巢，称重，计算子宫指数、卵巢指数；HE染色，光镜下观察子宫、卵巢组织形态学改变；采用放射免疫法测定血清生殖激素水平。结果各组大鼠子宫指数、卵巢指数之间差异无统计学意义；与正常组比较，模型组大鼠组织形态学异常，动情周期延长，血清生殖激素水平明显下降；与模型组比较，治疗组动情周期缩短，血清生殖激素水平明显升高；桃红组血清E2、P、LH、FSH明显升高；治疗组与桃红组比较动情周期缩短，血清E2、P明显升高。结论寒凝血瘀时内分泌紊乱，卵巢功能异常，加减温经汤可通过调节内分泌，改善卵巢功能而治疗寒凝血瘀型妇科疾病。Objective To observe the influence of Jiajianwenjing Decoction on rat ovarian of model with blood stasis due to cold stagnation and investigate the mechanism of Jiajianwenjing Decoction for treating gynecopathy of type of blood stasis due to cold stagnation. Methods To establish the rat model of blood stasis due to cold stagnation by putting rats into ice water （ 0 - 1 ℃ ）. The estrous cycles of rats were determined by vaginal smear, the weight of uterus, ootheca was measured and the uterus index and ovarian index were detected. After HE stain the histomorphological changes of rat uterus and ootheca were observed with the light microscope and the serum levels of generative hormones were detected with radioimmunoassay （ RIA ）. Results The difference of uterus index and ovarian index in different groups was insignificant statistically. In the model group the histomorphological changes were abnormal, the estrous cycle was prolonged and the serum levels of generative hormones decreased obviously compared with that of the normal group. In the treatment group the estrous cycle was shortened obviously and the serum levels of generative hormones increased compared with that of the model group. In the Taohong group the serum levels of estradiol （ E2 ） , P, luteinizing hormone （ LH ） and follicle - stimulating hormone （ FSH ） increased remarkably. The estrous cycle was shortened and the levels of E2 and P increased in the treatment group compared with that of the Taohong group. Conclusion The endocrine is disturbance and the ovarian function was abnormal in the gynecopathy of type of blood stasis due to cold stagnation. Jiajianwenjing Decoction can treat gynecopathy of type of blood stasis due to cold stagnation through regulating endocrine and improve the ovarian function.
