摘 要:目的探讨利用肝损伤早期和淤胆血清培养体系从大鼠骨髓细胞中筛选、诱导分化和扩增肝干细胞亚群,以快速、高效地获取骨髓源性肝干细胞。方法建立淤胆型肝损伤大鼠模型和制备含5%淤胆血清病理条件培养液,以促进骨髓源性肝干细胞的体内、外筛选和扩增,采用免疫细胞化学染色和RT-PCR方法检测培养2周时骨髓细胞各种肝干细胞标志表达。结果病理条件培养液培养2周的骨髓细胞形成肝细胞样集落,细胞表达胚胎期肝干细胞特征性蛋白。结论淤胆型肝损伤早期大鼠体内环境和含淤胆血清体外病理微环境可能有效地从骨髓细胞中筛选骨髓源性肝干细胞,从而为临床细胞移植治疗各种肝脏疾病提供种子细胞。Objective To explore the selection, differentiation and amplification of bone marrow-derived liver stem celIs(BDLSCs) from rats with early-stage hepatic injury in a culture system containing cholestatic serum, so as to harvest BDLSCs quickly and effectively. Methods BDLSCs were selected and amplified by setting up cholestatic hepatic injury models and preparing pathological condition media containing 5% cholestatic serum. Immunocytochem- istry and reverse transcription-polymerase chain reaction(RT-PCR) methods were applied to determine the expression of characteristics of liver stem cells. Results Bone marrow cells were able to form hepatocyte-like colonies in the presence of pathological condition media, and express characteristic proteins of fetal hepatocytes. Conclusion The environment of rats with early-stage hepatic injury in vivo and pathological microenvironment containing cholestatic serum in vitro may effectively select BDLSCs from bone marrow cells, which provides a useful resource for cell transplantation therapy in liver diseases.
