摘 要:HRP注入大鼠一侧终纹床核,存活48小时,再将1%Formalin导入胃内造成内脏伤害刺激,存活2小时后,灌注取材.对延髓切片进行HRP是色和抗FOS(ABC法)、抗TH(PAP法)免疫组化反应的三重反应.在光镜下发现“延髓内脏带”内出现七种阳性细胞,包括HRP阳性、TH和FOS免疫反应阳性;TH/HRP、FOS/HRP、FOS/TH等双重阳性以及FOS/HRP/TH三重反应阳性细胞.这些阳性细胞主要见于内脏带背内侧部的孤束核,其次是腹外侧区的腹外侧网状核(VLR),中间部较少.TH和FOS免疫阳性细胞以及TH/FOS双重阳性细胞为双侧分布,HRP/FOS、HRP/TH以及HRP/TH/FOS阳性细胞也为双侧分布但以注射区同侧为多。Horseradish peroxidase (HRP) was stereotaxically injected into the bed nucleus of the stria terminalis (BST) in the rat.Two days after, noxious visceral stimulation was given by injection formalin into the stomach under anesthesia of metoxyflurane gas. Two hours late, the rats were perfused and the brain was sectioned. The sections from the medulla were subjected to HRP reaction, immunohistochemical reactions for FOS protein (ABC method ) and tyrosine hydroxylase (TH, PAP method) successively. It is observed that expression of FOS protein in catecholaminergic neurous in medullary visceral zone (MVZ) (including vagus complex, ventrolateral medulla and a portion of reticular formation between them) project to the BST in rats. Seven kinds of labelled cells were found in medullary visceral zone. HRP retrograde labelled cells were seen in MVZ bilaterally with evident ipsilateral predominance, and TH-LI and FOS-LI single labelled cells were bilaterally distributed, HRP/TH-, TH/FOS double labelled neurons were most numerous than HRP/FOS ones. HRP/TH/FOS triple labelled neurous which were less in number were mainly distributed at the dorsomedial part,in the nucleus of tractus solitarii, and the ventrolateral part, ventrolateral reticular nucleus of MVZ, Whereas only a few labelled cells were located in its intermediate part. The significance of expression of FOS protein in TH neurons in MVZ, which project to BST in rats. receiving visceral noxious stimulation were discussed. The results showed that the nearons in MVZ and the bed nucleus of stria terminalis might be involved in stress response to noxious stimulation.
