液质联用方法测定鸡血浆中妥曲珠利及其代谢产物

摘　　要：初步建立肉鸡血浆中抗球虫药物妥曲珠利及其代谢产物妥曲珠利砜的液质联用（LC／MS）定性定量的测定方法。血浆样品加乙腈混匀后离心。取上清液进行液质联用（LC／MS）分析。色谱分析采用C18反相柱，流动相为乙腈-水溶液（体积比为55：45），质谱分析采用ESI离子源、二级串联质谱、负离子检测方式进行检测，妥曲珠利定量分析采用分子离子峰。该方法可以准确测定妥曲珠利的变化并判断代谢物的产生，在1次进样中实现同时检测妥曲珠利及其代谢物，妥曲珠利检测的线性范围为1—500ng·mL^-1，定量检测下限为0．5ng·mL^-1。此方法与液相色谱法相比灵敏度高、可靠、快速，该方法的建立为今后同类药物的定性定量分析提供基础。LC/MS was established to determine of toltrazuril and tohrazuril sulfone in chicken plasma. The plasma samples were mixed with acetonitrile, and spinned, then the supernatant was analysed by LC/MS. LC： C18 column , the mobile phase consisting of acetonitrile and water （ V： V = 55： 45 ）. MS ： electrospray ionization （ ESI）, second contacts the mass spectrum, negative ions analysis. Quantitative ion of toltrazuril was analysed with molecule ions peak. This method could analyse the change of tohrazuril and the metabolite accurately. The linear range of tohrazuril was 1 -500 ng ·mL^-1 and limit of detection was 0. 5 ng·mL^-1. This method is found to be more sensitive, credible and faster than HPLC. It can be used research of this kind of drugs in further.
