摘  要:目的观察川芎嗪对血管紧张素Ⅱ(angiotensinII,AngⅡ)诱导的血管平滑肌细胞(vascu lar smooth musc le cells,VSMCs)中钙调神经磷酸酶(Calc ineunrin,CaN)活性及原癌基因c-fos表达水平的影响。方法建立AngⅡ诱导VSMCs增殖模型,应用免疫细胞化学法观察不同浓度的川芎嗪在不同时段内对血管平滑肌细胞表达c-fos的影晌;并用酶促反应定磷法测定CaN活性变化。结果与正常对照组比较,AngⅡ组能够明显刺激VSMCs增殖,细胞增殖活度明显升高(P<0.01):AngⅡ作用后VSMCs中CaN活性和c-fos表达量(A值)较正常对照组均显著增高(P<0.01)。同时加川芎嗪作用后,各组CaN活性和c fos表达水平均显著下降(P<0.01)。结论AngⅡ能显著刺激血管平滑肌细胞增殖,川芎嗪能使血管平滑肌细胞中原癌基因c-fos的表达减弱及增殖受抑。Objective To study the effects of tetramethylpyrazine (TMP) on calcineurln (CaN) and C-fos gene expression in theproliferation of vascular smooth muscle cells (VSUCs) treated by Ang Ⅱ. Methods A cell proliferating model of VSMCs induced by angiotcnsinⅡ( AnG Ⅱ) was established; the effect of tetramethylpyrazine(TMP) on calcineurin(CaN) was detected by enzyme reaction phosphorus measurement ;the effect of etramethylpyrazine(TMP)on c -fos gene expression was observed by immunocytochemlcal stain and image analysis technology( A value). Results Cell proliferation activity, CaN activity and the expression levels of PCNA increased significantly in VSMCs proliferation induced by Ang Ⅱ ( P 〈 0.01 ). While treated with TMP, the index was obviously reduced compared with Angll group (P 〈 0.01 ). Conclusion The VSMCs proliferation induced by Ang Ⅱ can be inhibited by TMP significantly, and the inhibiting mechanism of TMP may be related to restraining the expression of c-fos in dose and time-dependent manner.
