摘 要:目的研究大鼠激光慢性高眼压视网膜损害相关基因表达谱的改变。方法采用5705点的大鼠高密度Oligo基因芯片,对运用532-二极管激光建立大鼠高眼压模型视网膜的基因表达情况进行检测,以对侧眼作为对照。结果通过筛选基因表达谱,得到5 277条在大鼠视网膜中有表达的基因,然后采用两两对比组合,从中筛选出了11条差异在2倍以上的基因,其中3条上调,8条下调。结论在大鼠高眼压模型视网膜中筛选到了11条表达差异>2倍的基因,可望为青光眼的神经损伤机制及治疗提供新的思路。Objective To systematically explore changes in gene expression in the retina of rats with laser-induced glaucoma and to validate the microarray data on eyes with experimental glaucoma. Methods Glaucoma was induced in the left eye of 25 Sprague- Dawley rats by repeated argon laser photocoagulation of the trabecular meshwork. The right eye served as the control. Retinas were isolated from glaucomatous and control eyes 35d after photocoagulation. Gene expression changes were analyzed by rat microarray chips which displayed a total of 5705 elements including Expression Sequence Tag (EST) clones, lOP was monitored daily by handheld tonometer, and retinas were collected 8d and 5 weeks after surgery. Results IOP elevation for 35d resulted in reproducible changes in levels of 11 mRNAs. In the glaueomatous retina, the number of upregulated genes was 3, and the number of downregulated genes was 8, respectively. The number of genes up- or downregulated was 0.33 % of all the genes examined. Conclusion Gene expression profiles in laser-induced glaucomatous rat retinas were determined, and only a very small population of genes was up- or downregulated in glaucomatous eyes. This result may be used to evaluate the neuroprotective potential of thera peutic agents that target this disease.
