摘  要:目的研究小剂量氢化可的松(HC)对内毒素诱导的脓毒症大鼠海马组织核因子-κB(NF-κB)、抑制因子κB(IκB)表达的影响。方法雄性Wistar大鼠54只随机分为:对照组(A组)6只,模型组(B组)24只、小剂量HC干预组(C组)24只。腹腔注射脂多糖(LPS)1 mg/kg建立脓毒症大鼠模型,尾静脉注射小剂量HC(6 mg/kg)作为干预,分别在注射后2、8、16、24 h分时段麻醉并灌注固定取脑。B组和C组每个时段点各6只。采用免疫组织化学方法及医学图像分析系统检测大鼠海马NF-κBI、κB的表达。结果模型组NF-κB表达较正常对照组显著上调(P<0.05),IκB表达先降低后逐步上升,较对照组亦明显增加(P<0.05),以8 h为最高;干预组海马NF-κB表达较模型组显著下调(P<0.05);IκB表达较模型组明显增加(P<0.05)。结论小剂量HC可通过诱生IκB来抑制NF-κB的表达,进而调控LPS诱导的脓毒症脑功能障碍,NF-κB信号转导途径在其发病机制中起重要作用。Objetive To investigate the effect of low-dose hydrocortisone (HC)on hippocampus nuclear factor kappa B (NF-kB), IkB expression in lipopolysaeeharide(LPS)indueed septic rats and the role of NF-kB signal transcriptiun pathway in pathogenesis. Methods Fifty-four rats were randomly divided into 3 groups: control group (A group, n=6), model group ( B group, n=24 ), low-dose HC treatment group (C group, n=24). The septic rat model was established by intmperitoneal injection LPS ( 1 mg/kg), as the intervention by caudal vein injection low-dose HC (6 mg/kg), each of B and C group was subdivied into 2,8,16,24 hours respectively after LPS injection (n=6). At serial time points,the animals in each group were sacrificed,brain tissue samples were harvested to determine NF-kB, IkB expression by imrnunhistochemistry in hippocampus. Results In B group: NF-kB expression was up regulated coanpared with A group(P〈0.05), IkB expression showed a down regulation firstly and escalated gradually before the peak of 8 hours time point (P 〈 0.05). NF-kB expression was in C group down regulated compared with B group ( P 〈 0.05 ) ; IkB expression was prominently enhanced (P 〈 0.05). Conclusions Low-dose HC inhibits the NF-kB expression by inducing the IkB expression, and then, to control the brain dysfunction of the septic rats, the NF-kB/IkB signal transcription pathway may play a very important role in the pathogenesis of the septic rat with brain dysfunction.
