摘  要:为了解天马颗粒剂的体内抗癌作用及其机理、毒性,建立人大肠癌SW-480细胞株肾被膜下移植瘤小鼠模型,设立天马颗粒剂治疗组、氟尿嘧啶(5-Fu)阳性对照组、联合用药组和安慰剂对照组。肿瘤细胞接种后第2天开始分别给药,连续用药8d后.采用病理形态学、免疫组化、图像分析技术等方法,定量检测各组移植瘤的细胞核形态及Ki-67、PCNA、wpp53和bcl-2的表达情况。各组抑瘤效应的强弱.同时对天马颗粒剂进行了毒理学研究。结果:(1)天马颗粒剂组肿瘤体积、重量均明显小于空白对照组和阳性对照组(P〈0.01),与联合用药组无显著性差异(P〉0.05);(2)天马颗粒剂和联合用药可改善移植瘤细胞及细胞核形态,并使核内染色质减少;(3)天马颗粒剂组和联合用药组的Ki-67表达和PCNA表达水平降低(P〈O.01);(4)天马颗粒剂能诱导SW-480细胞凋亡,下调wpp53。bcl-2表达}(5)天马颗粒剂安全无毒。For probing into the anticarcinomatous effects and toxicology of Tian Ma Granule in vivo the transplanted neoplasms model was established by injecting SW-480 cell strain of human colorectal carcinoma under renal capsule of mice,and four experimental groups were designed,including group of Tian Ma Granule treatment,group of 5-Fu treatment,group of combined treatment and placebo control group,comparatively studied. Drug treatment was started 2 days after carcinomatous cells implantation and lasted for 8 days. Then the nucleus morphology cells, the expressions of Ki-67,PCNA,wpp53 and bcl-2 in these cells, and the anticarcinomatous effect in respective groups were detected and compared ; the toxicological study on Tian Ma Granule was concurrently conducted. Pathological morphology, immunohistochemistry, image pattern analysis and quantitative determination were applied in the study. As results.(1)the size and weight of the neoplasms body in the Tian Ma Granule group were markedly less than that in the 5-Fu treatment group and in the placebo group( P〈0.01) ,while that in the Tian Ma Granule group were of no marked difference against that in the combined treatment group( P 〉0.05); (2)Tian Ma Granule treatment and combined treatment were able to improve the morphological structure of the implanted carcinomatous cells and their nucleus,and able to reduce the chromatin in the nucleus; (3)Tian Ma Granule treatment and combined treatment were able to reduce Ki-67 expression and the expression level of PCNA( P 〈0.01);(4) Tian Ma Granule treatment was able to induce SW-480 cell apoptosis and to down-regulate wpp53, bcl-2 expression;and (5)Tian Ma Granule was non-toxic and was safe.
