摘  要:【目的】观察重组人白细胞介素6(rIL-6)在体外对人近曲肾小管上皮细胞株(HK-2)增殖及生物学表型的影响。探讨rIL-6对HK-2细胞作用的时间-效应及剂量-效应关系。【方法】用MTT比色法检测rIL-6对细胞增殖的影响;用流式细胞术检测rIL-6对细胞周期及α-平滑肌肌动蛋白(α-SMA)阳性细胞百分率的影响;倒置显微镜观察rIL-6对HK-2细胞形态改变的影响。【结果】MTT比色法示,rIL-6在10~50ng/mL作用浓度范围内促进HK-2细胞增殖,呈剂量、时间依赖效应;rIL-6作用浓度为100~200ng/mL时抑制细胞增殖,随作用浓度增加抑制作用加强(F=201.582,P<0.001),(F=43.943,P<0.001)。rIL-6影响HK-2细胞的DNA合成。HK-2细胞有基础量的α-SMA表达,25ng/mLrIL-6与HK-2孵育1~48h后,α-SMA阳性细胞百分率高峰出现在48h(F=442.22,P<0.001)。rIL-6干预下,HK-2细胞形态由卵园型渐转变为长棱型、长条型。【结论】rIL-6可促进HK-2细胞增殖及生物学表型的转化,但大剂量的rIL-6可抑制HK-2细胞增殖。[Objective] The study was aimed to determine the in vivo proliferation and phenotype changes of human proximal tubular epithel/al cells (HK-2) responding to recommended human intedeukin-6-induced. [Methods] Cell proliferation was measured by MTT assay. The positive cells percentage of α-smooth muscle -actin (α-SMA) were analyzed by flow cytometry. The morphologic transforming of HK-2 cells was observed by light microscopy. [Results] Incubation for 48 hours with 10-50 ng/mL rIL-6 caused concentration and time-depondent stimulation of HK-2 cells proliferation, whereas rIL-6 inhibited HK-2 cells proliferation at 100-200 ng/mL respectively(F=201.582, P〈 0.001 ),(F=43.943,P〈 0.001). The DNA synthesis of HK-2 cells was influenced by rIL- 6. A basal level of α-SMA positive cells percentage existed in native HK-2 cells. Incubation for 1-48 hours with 25 ng/mL rIL-6 resulted in the maximal response at 48 hours(F=442.22, P〈 0.001). The HK-2 cells morphological transformation was observed after incubation with rIL-6. [Conclusion] The results indicate that rIL-6 could enhance HK-2 cell proliferation and contribute to HK-2 cell phenotype changes. In contrast, excessive rIL-6 could inhibit HK-2 cells proliferation. This could be a new discovered mechanism for how rIL-6 exhibits pathophysiological effects on tubular cells ultimately leading to tubulointerstitial fibrosis.
