复元愈肝胶囊对脂肪肝大鼠肝细胞凋亡的影响

摘　　要：目的:研究复元愈肝胶囊对脂肪肝大鼠的作用机理。方法:采用高脂饮食喂饲大鼠复制脂肪肝模型,同时用复元愈肝胶囊治疗,以东宝肝泰为对照药,观察各组动物的肝功能、血脂及肝组织学变化;运用流式细胞仪和免疫组化法检测大鼠肝细胞凋亡及Bc l-2、Bax蛋白表达。结果:与模型对照组相比,复元愈肝胶囊低、中、高剂量组能显著降低血脂,改善肝功能(P<0.01,P<0.05),明显优于东宝肝泰对照组(P<0.01,P<0.05)。模型对照组大鼠肝细胞凋亡程度明显高于各用药组及正常对照组(P<0.01);各用药组与模型对照组比较,肝细胞的凋亡率明显下降(P<0.05,P<0.01),Bc l-2基因蛋白表达量增高(P<0.05),Bax基因蛋白表达量降低(P<0.05)。结论:复元愈肝胶囊通过促进Bc l-2表达和抑制Bax表达,阻断脂肪肝大鼠肝细胞凋亡,而达到治疗脂肪肝的作用。Objective To investigate the effect of Capsule Fuyuan Yugan on rat fatty livers induced by high fat feeding. Methods Animal models were established by feeding high caloric diet and was administered Capsule Fuyuan Yugan or Dongbao Cantai as a control serum activities of liver - associated enzymes, blood lipids, liver triglyeerides and the liver histology of rats of all groups were assayed. Flow Cytometry and immunohistochemistry technique were used to detect the apoptosis of hepatocytes and the expression of Bel -2,Bax in hepatocytes, respectively. Results The levels of serum activities of a]anine aminotransferase （ ALT）, aspartate aminotransferase （ AST）, total cholesterol （ TC ）, trlglycerides （ TG ） and liver triglycerides in the different dose groups of Capsule Fuyuan Yugan were significantly lower than fatty liver model group and were similar to those of the normal group. The rate of apoptosis in treatment groups was significantly lower than that in fatty liver model group. The expression of Be] - 2 in treatment groups was significantly higher than that in fatty liver model group （ P 〈 0. 05 ） ,and the expression of Bax in treatment groups was significantly lower than that in fatty liver model group （ P 〈 0.05 ）. Conclusions Capsule Fuyuan Yugan might be effective in treating rat fatty liver induced by high fat feeding.
