摘 要:选用β-内酰胺酶量明显增加的耐药阴沟杆菌,用透析方法分析β-内酰胺酶与对酶稳定抗生素氨曲南和对酶不稳定抗生素头孢孟多相互作用过程,以研究酶介导的细菌耐药机制。结果表明,产酶株对头孢孟多的耐药以酶对药物的水解为主;对氨曲南的耐药依据β-内酰胺酶与氨曲南的不同作用过程而不同。可以是酶对药物“牵制”和缓慢水解同时并存,也可能是其它机制。Resistant mechanism of Enterobacter cloacae producing large amount of β-lactamase was studied by dialyzing the β-lactamase against a non-substrate antibiotic,aztreonam(AZ)and a substrate antibiotic,cefamandole(CMD)respectively,The activity of β-lactamase in dialysis bag and the concentration of antibiotic in the outer compartment were mesured by UV and microbial assay concomitantly.Results showed that the resistant mechanism of the organism to CMD was hydrolysis of the drug,while the resistant mechanism to AZ was depended upon the interaction between AZ and the β-lactamase,It could be‘ontained’and/or slowly hydrolyzed of AZ by β-lactamase or other mechanisms.
